【摘要】 目的 探究杜仲叶和栀子提取物对白色脂肪细胞褐变的促进作用，并探究其可能的机制。方法 采用CCK8法、Western blotting法、qRT-PCR法、油红O染色法、BODIPY染色法、线粒体荧光染料检测杜仲叶和栀子提取物对成熟的3T3-L1细胞褐变的影响，采用自噬激活剂(雷帕霉素)探明其可能的作用机制。结果 研究显示杜仲叶和栀子提取物能够上调褐变相关蛋白及mRNA(PRDM16、PGC-1α、UCP-1)的表达(P均lt; 0.05)，抑制自噬相关蛋白(Beclin-1、p62)和mRNA(LC3、Beclin-1、p62)及自噬小体的表达(P均lt; 0.05)，抑制脂滴积累，增加线粒体丰度。采用自噬激活剂(雷帕霉素)恢复杜仲叶和栀子提取物对3T3-L1细胞自噬水平后(P均lt; 0.05)，发现杜仲叶和栀子提取物对3T3-L1细胞的褐变作用也被自噬激活剂逆转(P均lt; 0.05)。结论 杜仲叶和栀子提取物可能通过抑制自噬进而促进白色脂肪细胞褐变，其在临床防治肥胖及相关代谢性疾病方面具有潜在作用。
