芍药内酯苷和芍药苷对环磷酰胺致血虚小鼠的补血作用及对GM—CSF,IL—3,TNF—α影响的比较研究(1)
[摘要]目的:比较研究芍药内酯苷、芍药苷对环磷酰胺致血虚小鼠的补血作用及机制。方法:采用环磷酰胺法复制小鼠血虚模型,灌胃芍药内酯苷、芍药苷单体提取物,检测小鼠外周血象、胸腺与脾脏指数及分离血清检验粒细胞-巨噬细胞集落刺激因子(GM-CSF)、白细胞介素-3(IL-3)、肿瘤坏死因子-α(TNF-α),比较二者的补血作用及机制。结果:芍药内酯苷30mg·kg-1与芍药苷30mg·kg-1均能升高白细胞数(P<0.01);芍药内酯苷30mg·kg-1与芍药苷30mg·kg-1均能升高红细胞数(P<0.01,P<0.001)。芍药内酯苷30mg·kg-1的胸腺指数明显升高(P<0.01),芍药苷30mg·kg-1的胸腺指数则无升高作用,二者相比,芍药内酯苷优于芍药苷(P<0.05);芍药内酯苷15mg·kg-1的胸腺指数明显升高(P<0.01),芍药苷15mg·kg-1的胸腺指数无升高作用,二者之间相比,无统计学差异。芍药内酯苷30mg·kg-1(P<0.01)、芍药内酯苷15mg·kg-1(P<0.05)、芍药苷30mg·kg-1(P<0.01)与芍药苷15mg·kg-1(P<0.05)均能升高GM-CSF的含量。芍药内酯苷30mg·kg-1与芍药苷30mg·kg-1均能升高IL-3(P<0.001)。芍药内酯苷30mg·kg-1与芍药苷30mg·kg-1均能降低TNF-α(P<0.01),二者相比,有明显统计学差异(P<0.01),芍药内酯苷优于芍药苷。结论:芍药内酯苷和芍药苷能够通过机体的免疫调节作用对抗环磷酰胺导致的血虚状态,且芍药内酯苷对造血负向调控因子的作用更强,与芍药苷协同发挥作用,是白芍养血柔肝功效的重要成分。

[Abstract]Objective: To compare the effects and mechanism of blood enriching on mouse model of blood deficiency syndrome induced by cyclophosphamide of albiflorin and paeoniflorin. Method: Albiflorin and paeoniflorin were determined by using animal models of blood deficiency syndrome induced by cyclophosphamide. The amount of WBC, RBC, HGB, index of thymus gland and spleen, and the changes of GM-CSF, IL-3 and TNF-α in serum were detected after the treatment. Result: Compared with the model group, the amount of WBC in the group of 30 mg·kg-1 albiflorin and 30 mg·kg-1 paeoniflorin were increased obviously(P<0.01). The amount of RBC in the group of 30 mg·kg-1 albiflorin and 30 mg·kg-1 paeoniflorin were increased obviously(P<0.01, P<0.001), which did not had a significant difference compared with the same dose. The index of thymus gland in the group of 30 mg·kg-1 albiflorin was superior to the model group(P<0.01), the difference was significant compared with the same dose of paeoniflorin(P<0.05). The GM-CSF in serum in all groups of 30 mg·kg-1 albiflorin , 15 mg·kg-1 albiflorin, 30 mg·kg-1paeoniflorin and 15 mg·kg-1 paeoniflorin increased obviously(P<0.01, P<0.05, P<0.01, P<0.05);The IL-3 in serum in both group of 30 mg·kg-1 albiflorin and 30 mg·kg-1 paeoniflorin also increased(P<0.001).The content of TNF-α in group of 30 mg·kg-1 albiflorin and 30 mg·kg-1 paeoniflorin were reduced(P<0.01), which showed the obvious difference compared with the same dose group(P<0.01). Conclusion: Albiflorin had the effect of blood enriching by regulating the immune function, same with the paeoniflorin.
