人胎盘底蜕膜间充质干细胞纹状体移植治疗帕金森病模型大鼠的实验研究(1)

     【摘要】目的研究人胎盘底蜕膜间充质干细胞(hPDBMSCs)纹状体移植对帕金森病(PD)模型大鼠行为学的影响。方法体外传代培养hPDBMSCs,使用6OHDA制备大鼠PD模型并随机分为移植组、假手术组、模型组,对移植组大鼠右侧纹状体进行hPDBMSCs移植,术后1 W、2 W、4 W进行行为学检测。免疫组化检测移植后1 W、2 W及4W各组大鼠损伤侧黑质酪氨酸羟化酶(TH)的表达。结果hPDBMSCs移植后2 W、4 W,移植组大鼠阿朴吗啡诱导的平均旋转圈数明显低于假手术组及模型组(P<0.01);免疫组化显示移植组损伤侧黑质部位移植后4 W TH阳性细胞数较移植前明显增加(P<0.01);与假手术组相比,移植术后1 W、2 W、4 W移植组TH阳性细胞明显增多(P<001)。结论hPDBMSCs移植能够抑制PD模型大鼠多巴胺能神经元丧失,改善PD模型大鼠的行为学症状,具有一定的治疗作用。

    【Abstract】ObjectiveTo investigate the effect of human placental decidua basalisderived mesenchymal stem cells(hPDBMSCs) striatum transplantation in the treatment of Parkinson’s disease(PD) on rat models.MethodshPDBMSCs were cultured in vitro.PD rat models were prepared by 6OHDA and divided into transplantation group,sham operation group and model group.HPDBMSCs transplantation was performed on the right striatum of the transplantation group,and behavioral detection was carried out 1W,2W and 4W after operation.Immunohistochemical method was used to detect expression of tyrosine hydroxylase (TH) in wounded sides among all the groups 1W,2W and 4W after transplantation.Results2W and 4W after transplantation,mean rotational circle induced by apomorphine(APO) in the transplantation group were lower than those in the sham operation group and the model group(P<0.01).Immunohistochemistry showed that TH positive cell numbers of the substantia nigra in the wounded sides 4W after transplantation significantly increased compared with those before transplantation in the transplantation group(P<0.01).Compared with the sham operation group,TH positive cell numbers 1W,2W and 4W after transplantation in the transplantation increased more significantly(P<0.01).ConclusionhPDBMSCs transplantation can inhibit the loss of dopaminergic neuron,improve behavioral symptoms of PD model rats,it has certain treatment effects.

    帕金森病(Parkinson’s Disease,PD)是一种较为常见的中老年人神经系统退行性疾病。目前临床上尚无有效方法延缓疾病的进展。随着干细胞技术的发展,研究人员开始探索通过细胞移植来延缓或抑制中脑黑质部多巴胺能神经元的凋亡,从而恢复神经环路中多巴胺递质的水平治疗PD。目前已有研究表明骨髓间充质干细胞(bone marrow mesenchymal stem cells,BMMSCs)移植能够改善帕金森病模型大鼠的行为学症状。人胎盘底蜕膜间充质干细胞(human placental decidua basalisderived mesenchymal stem cells,hPDBMSCs)具有和骨髓间充质干细胞相似的形态学、免疫表型和功能特点[1]。因而,本实验拟通过对PD大鼠纹状体进行hPDBMSCs移植,探讨人胎盘底蜕膜间充质干细胞移植对PD模型大鼠的行为学的影响。

    1資料与方法1.1材料

    1.1.1实验动物实验中所用大鼠均为SPF级SD大鼠,雌雄各半,共100只,体重180~220 g,8~12周龄,由广西医科大学实验动物中心提供(许可证号:SCXK桂2003-0005)。, http://www.100md.com(肖振勇 卢国辉 覃军 闫宪磊 陈家康 李学东)
