川崎病Sema4D/IL6轴介导中性粒细胞和内皮细胞相互作用的研究

【摘要】目的探讨信号素4D(Sema4D)/白细胞介素6(IL6)轴在川崎病(KD)中性粒细胞与内皮细胞间的相互作用。方法选取55例急性期KD患儿作为KD组,30例健康儿童作为HC组,ELISA法检测两组外周血Sema4D和IL6浓度,流式细胞术检测中性粒细胞表面Sema4D表达;Sema4D体外刺激内皮细胞并检测IL6表达;用内皮细胞条件培养基处理中性粒细胞后检测Sema4D脱落情况。
结果与健康儿童比较,急性期KD患儿外周血Sema4D和IL6明显升高,而Sema4Dhigh中性粒细胞明显降低,差异有统计学意义(Plt;0.05);与未刺激组比较,Sema4D刺激组的内皮细胞IL6表达显著增加,差异有统计学意义(Plt;0.05);与未刺激组相比,Sema4D处理组的内皮细胞条件培养基显著增加中性粒细胞Sema4D的脱落(Plt;0.01)。
结论川崎病冠脉病变过程中可能存在由Sema4D/IL6轴介导的中性粒细胞和内皮细胞互作方式。

【Abstract】ObjectiveTo explore the role of semaphorin 4D (Sema4D)/interleukin6 (IL6) axis in interaction between neutrophils and human coronary artery endothelial cells (HCAECs) in Kawasaki disease (KD).
MethodsA total of 55 patients with acute KD were selected as KD group, and 30 healthy children were selected as HC group.
