原花青素对氨基脲染毒小鼠免疫功能的影响

     【摘要】目的:探讨原花青素(PC)对氨基脲(SEM)染毒小鼠免疫功能的影响。方法:50只成年昆明(KM)种小鼠,随机分为5组:溶剂对照组、SEM染毒组、低、中高PC保护组。溶剂对照组灌胃去离子水; SEM染毒组,前4周灌胃去离子水,后2周灌胃56.25mg/kg·bwSEM溶液;低、中、高剂量PC保护组,先分别灌胃PC 100、200、400mg/kg.bw4周,继续分别灌胃同剂量SEM溶液2周。以上各组动物均用去离子水作溶剂,按10ml/kg.bw体积灌胃,连续灌胃6周,每日1次。末次灌胃24h后,采血后处死小鼠,摘取胸腺、脾脏等脏器检测相关指标。结果:低、中、高剂量PC保护组小鼠胸腺、脾脏脏器系数均分别高于SEM染毒组(p<0.05);经PC保护后,脾脏组织形态较SEM染毒组有明显改善,脾小体结构完整,红、白髓分界渐趋清楚,数目明显增多。脾索、脾窦排列趋于整齐,生发中心清楚可见;中、高剂量PC保护组NK细胞活性、MФ吞噬功能、AFC、SI均明显高于SEM染毒组(p<0.05);与SEM染毒组相比中、高剂量PC保护组SOD活性升高,MDA含量下降,差别均有统计学意义(p<0.05)。结论:一定剂量的PC可减轻SEM免疫损伤作用,其机制可能与增强机体抗氧化作用有关。

    【Abstract】Objective:To investigate the effect of procyanidins (PC) on the immune function of mice exposed by semicarbazide (SEM). Methods:mice were sacrificed after blood collection, the. 50 adult Kunming (KM) mice were randomly divided into 5 groups: solvent control group, SEM exposure group, low, medium and high doses of PC protection group. Solvent control group was intragastric administrated deionized water. SEM exposure group were intragastric administered 56.25 mg/kg.bw SEM solution in the first 2 weeks, then gavage deionized water for 4 weeks. Three doses of PC protection groups were fed PC 100, 200 and 400 mg/kg.bw separately in the first 4 weeks, and were fed the same dose of SEM solution for the next 2 weeks separately. All animals in each group were intragastric administration once a day according to 10 ml/kg.bw volume. After administered orally 6 weeks, mice were sacrificed, removal of the thymus, spleen and other organs, and detected related indicators of blood, tissues and organs. Result:The thymus and spleen organ coefficient in mice of low, medium and high doses of PC protection groups were, respectively, higher than the SEM exposure group (p <0.05). The spleen morphology of mice in three PC protection groups, which showed integrity spleen body structural, clear and increased boundaries between red and white pulp, neatly arranged splenic cord and sinus and clearly visible germinal center, were much better than SEM exposure group ......
