大肠埃希菌在炎症相关结直肠癌小鼠模型中对结直肠癌生长的作用(1)

     【摘 要】

    目的:本研究探讨大肠埃希菌在炎症相关结直肠癌小鼠模型中对肿瘤生长的作用。方法:实验雄性小鼠(C57BL/6)24只按体重大小平均随机分为两组:A组:给予氧化偶氮甲烷(Azoxymethane, AOM)联合葡聚糖硫酸钠(dextran sulfate sodium salt,DSS)制造炎症相关小鼠结直肠癌模型,B组:AOM、DSS给药方法同A组,其余时间饮用含大肠埃希菌的饮用水。均在第10周末处死小鼠,取得结直肠组织,计数肿瘤个数。结果:造模结束后A组中肿瘤计数总数为29,平均为2.90±2.183,B组总数为63,平均为9.00±3.555,两组差异显著(P<0.05)。A组成瘤率58.33%,B组成瘤率58.33%,病理均提示腺癌,两组间成瘤率无明显差异(P>0.05)。结论:大肠埃希菌在炎症相关性结直肠癌的小鼠模型中起促进肿瘤生长作用。

    E coli.promote tumorigenesis in a mouse model for colitis-associated cancer

    Abstract:

    Objective:Elevate the effection of E coli. for tumorigenesis in mouse model of colitis-associated colorectal cancer induced by azoxymethane (AOM) and dextran sulfate sodium salt(DSS). Methods: 24 male mice were randomly divided into group A and group B according to weight. The mice in Group A received intraperitoneal administration of AOM(10mg/kg)for one time, then drunk freely 1.5-2.1% DSS at the first, fourth, seventh week for three circles, the left time drunk freely distilled water. The mice in Group B received AOM and DSS as the same as group A, then drunk water including E coli. for left time.All mice were killed at the end of the tenth week.Collected the colorectum from anus to the end of cecum, count the number of tumor. Results: The number of tumor is 29,and 2.90±2.183 averagely in group A, and 63 in total , 9.00±3.555 averagely in group B.There is difference between the two groups(P<0.05) , The tumor formation rate is no difference (P>0.05), and 58.33% in group A, 58.33% in group B. Conclusion:E coli. promote tumorigenesis in mouse model for colitis-associated cancer.

    前言

    结直肠癌是常见的恶性肿瘤,针对结直肠癌的发生、发展的相关研究越来越深,从分子生物学到基因学,其发生机制逐步被发现,其发生相关因素包括遗传 [1]、高牛肉饮食 [2]及炎症性肠病 [3]等等。此外,结直肠癌的发生、发展还与肠道微生态失调有关,有研究指出无菌环境中生长的小鼠发生结直肠癌几率增高,大肠埃希菌属及具核梭菌属含量在结肠癌组织中及结肠癌患者大便中的含量明顯高于正常结肠组织及正常人大便[4-5]。本实验探讨大肠埃希菌在炎症相关结直肠癌小鼠模型中对结直肠肿瘤发生、发展的影响。, 百拇医药(王磊 时军利 李炳庆)
