应用G-CSF动员骨髓干细胞治疗重型颅脑损伤的研究(1)

     【摘要】探究重组人粒细胞集落刺激因子(rh-G-CSF)治疗重型颅脑损伤的有效性及临床应用安全性。方法将60例重型颅脑损伤患者,随机分成实验组30人,对照组30人。实验组连续3天给予G-CSF 300μg/天。应用流式细胞仪监测并比较治疗前后外周血中CD34+和CD133+细胞计数及磁共振波普分析(MRS)评价治疗前后受损部位的N-乙酰氨基酸(NAA)含量,常规评价其功能恢复情况。监测外周血白细胞计数评价其安全性。结果实验组治疗后CD34+、CD133+细胞计数和受损部位的NAA含量较治疗前明显增加(p<0.05),白细胞计数同步增加(P<0.05)。治疗1个月时实验组GCS评分较对照组略有改善,但差异无显著性(t=0.032,P>0.05),随访3个月、6个月时GCS评分较对照组均明显升高(t=1.418,1.592,P<0.05)且随访6个月时实验组的疗效明显优于对照组(Z =3.576,P<0.01),实验组未出现严重不良反应。结论用G-CSF动员骨髓干细胞迁移至脑损伤区域并分化为神经细胞,明显改善神经功能。

    【ABSTRACT】The aim of this study was to assess the safety and efficacy of treatment for patients with severe traumatic brain injury (sTBI) with transplantation of granulocyte colony stimulating factor(G-CSF) -mobilized autologous bone marrow stem cells (BMSCs) MethodsSixty patients with sTBI were randomly divided into the control group (group A),the group treated with transplantation of G-CSF -mobilized BMSCs (group B,G-CSF ,300μg/day ×3 days).ResultsCount of CD 34+ and CD 133+ cells in peripheral blood. was observed by flow cytometry.
