慢性阻塞性肺疾病大鼠Toll样受体4表达的变化及其对氨溴索反应性的研究(1)

【摘要】目的 观察大鼠烟雾暴露慢性阻塞性肺疾病(COPD)模型支气管肺泡灌洗液TLR4的表达，观察氨澳索的干扰作用，以探讨TLR4在COPD发病机制中的作用及氨浪索对其的影响。方法实验大鼠随机分为 3组：正常对照组 (n =8)，COPD模型组 (n =10)，氨溴索干预组 (n =10)。应用酶联免疫吸附测定(ELISA)法测定各组大鼠肺泡灌洗液中TLR4的浓度。结果 COPD模型组大鼠支气管肺泡灌洗液中TLR4表达强度较正常对照组明显降低(P<0.01)；氨溴索干预组支气管肺泡灌洗液中TLR4水平较COPD模型组显著升高(P<0.01)，但与正常对照组相比仍有显著性差异 (P<0.05)。结论慢性阻塞性肺疾病大鼠肺泡灌洗液中TLR4表达水平较正常组低下，这可能与COPD慢性气道炎症的形成密切相关；长期应用氨溴索能上调其表达，提示氨溴索对COPD大鼠模型气道炎症有明显的抑制作用。

【Abstract】 Objective To investigate the expressionof Toll-like receptor 4 (TLR4) in smoke-induced of chronic obstructire pulmonary disease(COPD) rat models，and to explore the role of TLR4 in the pathogenesis of COPD and the effeets of ambroxol on it. Methods 28 male wistar rats were randomly divided into three groups(Group A：normal control group，group B：model group，Group C：ambroxol treated group). .The concentrations of TLR4 were determined by enzyme linked immunosorbent assay.Results The expressions of TLR4 in COPD model group were significantly decreased than those in normal control group (p<0.0l).The expressions of TLR4 in ambroxol treated group were increased than those in COPD model group(p
