奥曲肽联合5-氟尿嘧啶对Hepa1-6肝癌细胞PAK1表达的影响(1)

     [摘要]目的研究不同浓度奥曲肽(OCT)及OCT联合5-氟尿嘧啶(5-Fu)对Hepa1-6肝癌细胞的作用效果,并探讨其可能的作用机制。方法 采用MTT法检测不同浓度OCT及OCT联合5-Fu对细胞活性的影响;采用逆转录聚合酶链反应(RT-PCR)检测PAK1mRNA的表达;细胞划痕愈合实验测定细胞扩散运动能力。结果 不同浓度OCT及OCT联合5-Fu组细胞生长抑制率均随剂量的增加而增加,与对照组相比有显著差异(P<0.01),且联合用药组均明显高于相应浓度OCT及5-Fu组(P均<0.05);OCT10mg/L联合5-Fu组较肝癌细胞组、OCT10mg/L组PAK1mRNA的表达明显下调(P均<0.01),但仍较正常肝细胞组高;OCT10mg/L联合5-Fu组较肝癌细胞组、OCT10mg/L组细胞移行运动能力显著减弱。结论 OCT10mg/L联合5-Fu为抑瘤最佳药物浓度,其主要通过减少PAK1的表达、减弱细胞移行运动能力而发挥抑瘤作用。

    The effect of combinating octreotide acetate and 5-Fluorouracil for the expression of PA

    [Abstract]Objective To research the effect of different concentration of Octreotide Acetate(OCT)and OCT combinate 5-Fluorouracil(5-FU) on hepatoma(Hepa1-6) cell,and to explore its role in the mechanism.Methods MTT method was used to test the activity of the cell acted by assay of different concentrations OCT and OCT combinate 5-Fu;the expression of PAK1mRNA was detected by reverse transcription polymerase chain reaction(RT-PCR);monolayer wound healing assay was used to detect Hepa1-6 cell motility.Results The inhibitory rate of cell growth in different concentrations of OCT and OCT combined with 5-Fu group increased with OCT dosage augment,and was significantly higher than that in controls(P<0.01),and the combined treatment group were significantly higher than the corresponding concentration of OCT group and 5-Fu group(P<0.05);The relative level of PAK1mRNA in OCT 10mg/L combined with 5-Fu group was significantly decreased compared with hepatocellular carcinoma group and OCT 10mg/L group(P<0.01),but still higher than normal group; the cell migration ability of OCT 10mg/L combined with 5-Fu group was weaken significantly compared with hepatocellular carcinoma group and OCT 10mg/L group ......
