兔椎间盘退变过程中软骨终板TNF-α表达及作用机理探讨(1)

     [摘要] 目的 研究兔椎间盘退变过程中软骨终板TNF-α表达及其对蛋白多糖、胶原合成的影响。方法 选用新西兰白兔36只,随机分为实验组和对照组。采用椎间失稳方式诱发椎间盘退变。于术后12w、24w和36w取材进行组织学观察、TNF-α含量检测。并采用来源猪的终板软骨细胞进行传代培养,采用同位素方法,检测外源性TNF-α对培养软骨终板细胞蛋白多糖及胶原合成的影响。结果 随着不稳时间延长,软骨终板出现明显退变,软骨终板变薄、钙化,细胞数目减少。实验组TNF-α含量较对照组明显增高,但24w与36w之间无差异。TNF-α处理组,培养终板软骨细胞的蛋白多糖合成和胶原合成量均较对照组显著性减少。结论 软骨终板退变过程中,早期TNF-α表达显著性增加,而TNF-α具有抑制培养软骨细胞蛋白多糖和胶原合成的作用。独活寄生汤对椎间盘退变炎性因子表达的影响可能是今后研究的方向。

    [Abstract] Objective To study the expression of TNF-αin endplate cartilage during the degenerative progress of rabbit spine, and the effect of TNF-α on synthesis of proteoglycan and collagen of cultured endplate cartilage cells.Methods thirty-six New Zealand rabbits were divided randomly into two groups,experimental group and control group.In the experimental group,the disc degeneration was created by intervertebral instability in rabbits.On the interval of 12w,24w,36w,the endplate cartilages were harvested to be examined histologically and the expression of TNF-α.The endplate cartilage cells cultured from adult pigs were used for detection of proteoglycan and collagen synthesis after treatment of exogenous TNF-α.Results With the expanding period of intervertebral instability,the endplate cartilage showed more degenerative changes including thinning and calcification of endplate and scarcity of cartilage cells.The contain of TNF-αin the experimental group was significantly higher than that of control group, but no significant difference was found between the experiment groups in 24w and 36w ......
