HAC70-多肽复合物疫苗对实验性肿瘤细胞抗肿瘤活性的研究(1)

    【摘要】目的:观察从艾氏腹水瘤细胞中提取的热休克蛋白70-抗原肽复合物(HAC70)诱导的抗肿瘤免疫效应,探讨其抗肿瘤机制。方法:提取热休克后的小鼠艾氏腹水瘤细胞内热休克蛋白70(HSP70)经肝素-琼脂糖亲和层析纯化,制成热休克蛋白-肽复合物疫苗,以体内免疫法检测HAC70的抑瘤作用,3H-TdR掺入释放法检测脾细胞对肿瘤细胞的抗肿瘤活性。结果:经HAC70免疫的荷瘤小鼠皮下肿瘤结节平均重(1.26±0.4)g,较对照组(3.14±0.56)g明显减轻(P<0.05);当效靶比为200:1时,免疫组脾细胞杀伤活性为(49.25±11.25)%,显著高于对照组[(18.50±9.95)%,P<0.01]及荷瘤组[(25.75±8.38)%,P<0.01]。结论:经肝素-琼脂糖亲和层析纯化的HAC70可通过诱导机体产生特异性杀伤艾氏腹水瘤细胞的活性,明显抑制荷瘤小鼠艾氏腹水瘤的生长。

    【Abstract】 Objective:To investigate the anti-tumor immuno-efficacy induced by HAC70 derived from Ehrlich ascites tumor cell and to study its mechanisms.Methods:After the heat-shock HSP70 from the Ehrlick ascites tumor cell was isilated and pureed by Heparin-Sepharose chromatography and got the HAC70 vaccine.The anti-tumor efficacy of splenocyte was determined by the 3H-TdR release assay.Results:The average tumor weight of the mice received HAC70 immunization was only(1.26±0.4)g,obvious difference(P<0.05),observed when contrasted with the control group(3.14±0.56)g.It was showed that specific cytotoxitic rate of splenocytes from the immunized mice to the Ehrlich ascites tumor cells was(49.75±8.93)%,significantly higher than from the Tumor-bearing(18.5±9.95)% and control group(25.75±8.38)%,P<0.01.Conclusion:HAC70 pureed by Heparin-Sepharose chromatography have the activities of inflicting the Ehrlick ascites tumor cell and inhibit obviously growth of the Ehrlick ascites tumor.

    对热休克蛋白70-多肽复合物疫苗(HAC70)抗肿瘤作用的研究表明HAC70在诱导荷瘤机体产生抗肿瘤免疫效应的过程中,热休克蛋白(HSPs)的基因并没有突变,是伴随着HSPs分子上的抗原肽引起的免疫反应的特异性导致的抗肿瘤效应,比较重要的有HSP70、HSP90、GP96等。

    本实验采用从艾氏腹水瘤细胞中提取的HSP70免疫BALB/c小鼠,制备热休克蛋白70-多肽复合物疫苗(HAC70)[1],观察其抗肿瘤作用,并以3H-TdR释放法检测HAC70诱导机体产生的特异性杀伤活性,报告如下:
