藏药镰形棘豆鼠李柠檬素对免疫抑制小鼠非特异性免疫功能的影响(1)

     摘要:目的 探讨藏药镰形棘豆提取物鼠李柠檬素增强免疫的作用机制。方法 选用昆明种小鼠,随机分为空白组、模型组、阳性药组及中药高、中、低剂量组,各给药组给予相应药物灌胃,连续10 d。第8日起,除空白组腹腔注射生理盐水外,其他各组均腹腔注射环磷酰胺,连续3 d。通过巨噬细胞炭粒廓清试验、巨噬细胞吞噬鸡红细胞试验、免疫抑制小鼠器官指数测定,观察鼠李柠檬素对免疫抑制小鼠非特异性免疫功能的影响。结果 与模型组和空白组比较,中药各剂量组能提高免疫抑制小鼠的吞噬活性、廓清指数和吞噬系数(P<0.05),增加免疫器官脏器指数;与阳性药组比较,中药各剂量组药效较低。结论 藏药镰形棘豆鼠李柠檬素具有一定的免疫增强作用,可促进小鼠鸡红细胞免疫后脾细胞血清溶血素的形成。

    Abstract:Objective To study action mechanism of rhamnocitrin in Oxytropis Falcata on enhance the immune function. Methods Kunming mice were randomly divided into blank control group, model group, positive control group, traditional Chinese medicine high, medium, and low dose groups. All treatment groups received gavage with relevant medicine for 10 days. From the 8th day, blank control group received intraperitoneal injection with normal saline, while other groups received intraperitoneal injection with cyclophosphamide for 3 days. Through macrophages carbon clearance test, macrophage phagocytosis of chicken erythrocytes test, and immunosuppressive mice organ index detection, effects of rhamnocitrin in Oxytropis Falcata on nonspecific immune function of immunosuppressive mice were observed. Results Compared with model group and blank control group, traditional Chinese medicine high, medium, and low dose groups could improve the phagocytic activity of immunosuppressive mice, clearance index, and phagocytic coefficient (P<0.05), and increase the organ index of the thymus and spleen. Compared with the positive control group, the efficacy of traditional Chinese medicine high, medium, and low dose groups was low. Conclusion Rhamnocitrin in Oxytropis Falcata can enhance the immune function, improve the formation of spleen cells of mice serum hemolysin of chicken red blood cell immune.

    镰形棘豆来源于藏药豆科植物镰形棘豆Oxytropis falcata Bunge和轮叶棘豆O.chiliophylla Royle的干燥全草,具有清热解毒、生肌愈疮、涩脉止血、通便等作用。主要用于治疗中毒病、黄水病、便秘、炭疽、疮疖痈肿等疾病,是藏医“三抗炎药”之一,收载于《晶珠本草》及《中华人民共和国卫生部药品标准(藏药)》(第一册)[1]。研究表明,镰形棘豆中含有黄酮类、生物碱类等多种活性成分,具有抗炎、抗菌、抗肿瘤等药理作用[2-7]。目前对镰形棘豆的抗炎作用研究较多,主要为黄酮类成分的抗炎作用。为了进一步确定该药材抗炎作用的有效活性成分,本实验采用提取、酸化、柱层析等方法制备了镰形棘豆黄酮中的主要成分鼠李柠檬素,观察镰形棘豆提取物鼠李柠檬素对免疫抑制小鼠的非特异性免疫功能的影响,为镰形棘豆黄酮的药效活性物质的利用及以后的剂型改革和临床用药提供多方面的实验依据。

    1 实验材料

    1.1 动物

    SPF级昆明种小鼠,8周龄,雌雄各半,体质量20~22 g,购于兰州大学实验动物中心,许可证号SCXK(甘)2009-0004。实验前,所有小鼠被分组放入塑料鼠笼,适应性饲养7 d(温度23~25 ℃,湿度50%~60%),喂食普通鼠饲料和蒸馏水。, http://www.100md.com(郭敏 张丽 姜华)
