养阴益气合剂对博来霉素所致肺纤维化大鼠的影响(1)

     摘要:目的 观察养阴益气合剂对博来霉素所致肺纤维化大鼠的影响,探讨其可能的作用机制。方法 将SPF级SD大鼠随机分为正常组、模型组、阳性药组和养阴益气合剂低、中、高剂量组。采用气管内滴注博来霉素方法复制肺纤维化大鼠模型,造模后1 d,各给药组给予相应药物灌胃。分别于第7、14、28日取肺组织,Masson染色观察病理变化,碱水解法检测羟脯氨酸(HYP)含量,Western blot检测基质金属蛋白酶-9 (MMP-9)和基质金属蛋白酶抑制剂-1(TIMP-1)蛋白表达。结果 造模7 d,病理切片显示模型组大鼠肺部胶原较正常组明显增多,即造模成功;HYP含量同步显著升高(P<0.01);造模28 d,病理切片显示模型组肺泡间隙胶原大量沉积,部分肺泡结构丧失,HYP含量仍处于高值(P<0.01)。经养阴益气合剂中、高剂量28 d干预治疗,肺组织胶原沉积均有所改善,且表现出剂量相关性,HYP含量明显减少(P<0.01),养阴益气合剂高剂量组表现出与阳性药组相当的效果。Western blot检测结果显示,造模7 d在正常肺组织低表达的MMP-9和TIMP-1蛋白水平显著升高(P<0.05),至28 d持续处于高表达状态(P<0.05)。与模型组比较,第14日养阴益气合剂高剂量组MMP-9水平明显降低(P<0.05),第7、14、28日TIMP-1表达水平明显降低(P<0.05)。结论 养阴益气合剂能有效抗肺纤维化,推断其通过抑制TIMP-1和MMP-9的表达,调控促进胶原的水解,抑制胶原蛋白的沉积,最终抑制肺纤维化。

    Abstract:Objective To investigate the effects of Yangyin Yiqi Mixture on pulmonary fibrosis caused by bleomycin in rats;To discuss its possible mechanism. Methods SD rats were randomly divided into normal control group, model group, dexamethasone acetate group, low-dose, middle-dose and high-dose Yangyin Yiqi Mixture groups. Bleomycin was used to establish pulmonary fibrosis rat models through endotracheal instillation. One day after molding, each medication group received gavage with relevant medicine. Lung tissues were taken on the 7th, 14th and 28th day. Masson staining was used to observe pathological change;alkalinehydrolysis was used to detect HYP content;Western blot was used to detect the protein expressions of MMP-2 and TIMP-1. Results Compared with the normal control group, on 7th day, collagen in lung tissue of rats in model group markedly increased;the content of HYP in lung tissue significantly increased (P<0.01).
