荆芥挥发油的提取及胶体磨包合工艺研究(1)

     摘要:目的 优选荆芥挥发油的提取及β-环糊精胶体磨包合工艺。方法 采用水蒸馏法进行提取,以浸泡与否、加水倍量、蒸馏时间为考察因素,挥发油体积为评价指标,考察荆芥挥发油的提取工艺;以β-环糊精与挥发油比例、加水倍量、研磨时间为考察因素,挥发油包合率为评价指标,采用胶体磨研磨法,正交试验优选包合工艺。采用紫外分光光度法和显微成像分析对包合物进行验证。结果 优选的提取工艺为荆芥加8倍量水,提取6 h;包合工艺为挥发油与β-环糊精比例为1∶8(mL∶g),加8倍量水,研磨30 min。结论 所选工艺合理、可行,可用于提取荆芥中挥发油及挥发油与β-环糊精的包合。

    Extraction Technology of Volatile Oil from Schizonepetae Herba and Inclusion Technology by Colloid Mill YANG Chun-jing, NI Jian, CAO Sa-li, XUE Dan, ZHANG Xin, XIA Zhen-wen, ZHAO Yang, YIN Xing-bin (Beijing University of Chinese Medicine, Beijing 100102, China)

    Abstract:Objective To optimize the extraction technology of volatile oil from Schizonepetae Herba and the inclusion technology by colloid mill of β-cyclodextrin. Methods Water distillation method was used to extract. Infusion or not, amount of water and distillation time were set as factors, and the volume of volatile was set as indexes to examine the extraction technology of volatile oil from Schizonepetae Herba;the ratio of β-cyclodextrin to volatile oil, inclusionn time, amount of water were set as factors, and the yield of inclusion compounds and inclusion rate of volatile oil were set as indexes to use colloid mill grinding method and orthogonal test to optimize the inclusion technology. Ultraviolet spectrophotometry and microscopic imaging analysis were used to verify this inclusion compound. Results The optimized extraction conditions were:adding 8-fold water, extracting for 6 h without infusion. The optimized inclusion conditions were:the proportion of volatile oil to β-cyclodextrin=1∶8 (mL∶g), adding 8-fold water, milling for 30 min. Conclusion The technology is reasonable and feasible, which can be used for the extraction of volatile oil from Schizonepetae Herba and the inclusion of β-cyclodextrin.

    荆芥为唇形科植物荆芥Schizonepeta tenuifolia Briq.的干燥地上部分,其功效为解表散风、透疹、消疮,用于感冒、头痛、麻疹、风疹、疮疡初起[1]。荆芥

    中的挥发油为荆芥的主要有效部位群之一[2-3],其中又以薄荷酮和胡薄荷酮为主要成分[4]。药理实验显示,其具有一定抗流感病毒、抗炎作用,对流感病毒感染小鼠有保护作用,可明显抑制二甲苯所致大鼠耳廓肿胀[5-6]。已有荆芥挥发油β-环糊精包合研究报道,但多采用饱和水溶液法[7-10]。胶体磨研磨法具有效率高、包合率高、适宜工业化大生产等优点[11]。本研究采用该法对荆芥挥发油进行包合,以期为进一步开发、利用荆芥药材资源提供参考。
