柳茶总三萜提取富集工艺优化及抗缺氧活性研究(1)
摘要:目的 优选藏药柳茶总三萜的提取富集工艺并研究其抗缺氧活性。方法 通过小鼠常压密闭缺氧实验筛选柳茶总三萜的提取溶剂;再以总三萜含量、提取率为评价指标,采用正交试验考察提取时间、提取次数、料液比,优选柳茶总三萜的提取工艺;然后考察大孔吸附树脂类型、上样浓度、洗脱剂浓度、洗脱剂体积,优选其富集工艺;最后通过小鼠常压密闭缺氧实验和急性减压缺氧实验研究其抗缺氧活性。结果 优选的提取工艺为:70%乙醇提取,料液比为1∶20,提取2次,每次1.5 h,平均总三萜含量可达32.25%;优选的富集工艺为:AB-8型大孔吸附树脂,上样浓度为2 mg/mL,80%乙醇5 BV洗脱,总三萜含量可达61.09%;柳茶总三萜抗缺氧活性较强,且存在一定的剂量依赖性。结论 本研究优选的柳茶总三萜提取富集工艺稳定、重现性好,所得柳茶总三萜具有显著的抗缺氧活性。关键词:柳茶;总三萜;提取工艺;富集工艺;抗缺氧
