参苓白术散对非酒精性脂肪性肝病模型大鼠瘦素及胰岛素抵抗的影响(1)
摘要:目的 观察参苓白术散对非酒精性脂肪性肝病(NAFLD)模型大鼠血脂、瘦素和胰岛素抵抗的影响,探讨其相关作用机制。方法 20只实验大鼠随机分为正常组5只(正常饲料喂养)和高脂饮食组15只,高脂饮食组大鼠给予高脂饲料喂养制作NAFLD模型,将成模大鼠随机分为模型组、参苓白术散组和吡格列酮组,每组5只,正常组和模型组给予生理盐水灌胃,参苓白术散组给予参苓白术散水煎液灌胃,吡格列酮組给予吡格列酮混悬液灌胃。药物干预8周后进行口服葡萄糖耐量实验,检测大鼠糖耐量变化及血清转氨酶、血脂、瘦素与胰岛素水平,并计算胰岛素抵抗指数。结果 与正常组比较,模型组大鼠血清转氨酶、血脂、瘦素、胰岛素、血糖和胰岛素抵抗指数均显著升高(P<0.05);与模型组比较,参苓白术散组和吡格列酮组大鼠血清转氨酶、血脂、瘦素、胰岛素、血糖和胰岛素抵抗指数均显著降低(P<0.05)。结论 参苓白术散通过降低模型大鼠转氨酶与血脂水平发挥治疗NAFLD作用,其机制可能与调节瘦素、改善胰岛素抵抗有关。

Abstract:Objective To observe the effects of Shenling Baizhu Powder on blood lipid, leptin and insulin resistance of non-alcoholic fatty liver disease (NAFLD) rats; To explore its mechanism. MethodsTwenty rats were randomly divided into normal group (normal diet feed, n=5) and high-fatdiet group (high-fat diet feed, n=15). Rats of high-fat diet group were fed with high-fat diet to establish NAFLD model. Model rats were divided into model group, Shenling Baizhu Powder group and pioglitazone group, with 5 rats in each group. Normal group and model group were given normal saline for gavage, while Shenling Baizhu Powder group and pioglitazone group were given Shenling Baizhu Powder and pioglitazone suspension by intragastric administration respectively. After 8 weeks of medicine intervention, all rats were subjected to oral glucose tolerance test. Serum transaminase, blood lipid, leptin and insulin levels were detected by biochemical analysis. Insulin resistance index was calculated. Results Compared with the normal group, serum transaminase, blood lipid, leptin, insulin, blood glucose and insulin resistance index increased significantly in the model group (P<0.05). Compared with the model group, serum transaminase, blood lipid, leptin, insulin, blood glucose and insulin resistance index in Shenling Baizhu Powder and pioglitazone group decreased significantly (P<0.05). Conclusion Shenling Baizhu Powder can conduct treatment of NAFLD by reducing the effects of transaminase and lipid levels in model rats, and the mechanism may be related to the regulation of leptin and the improvement of insulin resistance., 百拇医药(王萌 张会存 刘欣 汪红兵)
