发酵前后不同分子量段全蝎白术混合品体外抗肿瘤研究(1)

     摘要:目的 研究全蝎白术混合发酵品的抗肿瘤活性,确定药物抗肿瘤的活性部位。方法 依据双指标优化提取工艺的最优条件对全蝎白术混合发酵品进行超声提取。采用超滤手段,分别用截留量为5 kD和10 kD的超滤膜对水提液进行超滤分段,测定各分子量段中蛋白质和多糖的含量,采用MTT法检测体外抗肿瘤活性,并以未发酵品做对照。结果 各分子量段对2种肿瘤细胞抑制效果具有差异性,醇沉上清液对肿瘤细胞的抑制效果最好,其次为分子量段>10 kD全蝎白术混合发酵品,且全蝎白术混合发酵品对肿瘤细胞抑制作用显著优于未发酵品。结论 全蝎白术混合发酵品对乳腺癌细胞、喉癌细胞均有较高的抑制作用,其活性部位主要为醇沉上清液和分子量段>10 kD全蝎白术混合发酵品。

    Abstract: Objective To study the antitumor activity of Scorpio and Atractylodis Macrocephalae Rhizoma mixed fermentation product; To determine the antitumor active sites of Scorpio and Atractylodis Macrocephalae Rhizoma mixed fermentation product. Methods Scorpio and Atractylodis Macrocephalae Rhizoma mixed fermentation product was extracted by ultrasonic assay on the basis of the optimal conditions of the double index optimization extraction process. The water extract was segmented by ultrafiltration membrane with a retention of 5 kD and 10 kD respectively. The contents of protein and polysaccharide in each molecular weight segment were determined. MTT assay was used to study the antitumor activity in vitro. The unfermented product was used as the control group. Results The inhibitory effects of different molecular weight segment on the two tumor cells were different, and the inhibitory effects of alcohol supernatant on the tumor cells were the best, followed by the SAHFP of molecular weight >10 kD segment. Scorpio and Atractylodis Macrocephalae Rhizoma mixed fermentation product was significantly better than that of unfermented products. Conclusion Scorpio and Atractylodis Macrocephalae Rhizoma mixed fermentation product has a high inhibitory effect on breast cancer cells and laryngeal cancer cells. The active sites of antitumor drugs are mainly the alcohol supernatant and the Scorpio and Atractylodis Macrocephalae Rhizoma mixed fermentation product of molecular weight >10 kD segment.

    全蝎為钳蝎科动物东亚钳蝎Buthus martensii Karsch的干燥体[1]143,主要活性成分为蝎毒,其毒性强烈,若服用不当,会出现一系列不良反应。因此,临床使用时需进行合理的药物配伍,进而降低全蝎的毒性,最大程度发挥其药效。《医宗金鉴》认为可用全蝎、僵蚕为醒脾汤化痰治标,更用人参、茯苓、白术温补中气,恢复脾气健运,助化痰祛邪,使邪去而不伤正。白术为菊科植物白术Atractylodes macrocephala Koidz.的干燥根茎,具有健脾益气、燥湿利水功效[1]103。王彦多等[2]将攻毒散结的全蝎与健脾益气的白术相配伍,发现球孢白僵菌发酵的全蝎白术混合发酵品(SAHFP)提取液具有显著的抗肿瘤作用。据此,本实验对有效部位进行探讨,以期通过超滤分段分离SAHFP中有效成分,进一步明确抗肿瘤活性部位,为该药物成分及其他中药配伍研究提供参考。, 百拇医药(毛会秀 陈美琦 刘金虎 吕迎兰 方园 张瑞 王集会)
