肺纤方对博来霉素诱导肺纤维化大鼠wnt3a、β-catenin、WIF1和LRP表达的影响(1)
摘要:目的 观察肺纤方对博来霉素诱导的肺纤维化大鼠wnt3a、β-catenin、WIF1、LRP表达的影响,探讨其抗肺纤维化的作用机制。方法 采用气管插管滴注博来霉素制作肺纤维化大鼠模型。将120只雄性Wistar大鼠随机分为假手术组、模型组、泼尼松组和肺纤方高、中、低剂量组,造模次日给予相应药物灌胃。于造模后第14、28日分2批进行动物处理,Western blot检测wnt3a、β-catenin、WIF1的蛋白表达,RT-PCR检测LRP 的mRNA表达。结果 与假手术组比较,第14、28日模型组大鼠肺组织wnt3a、β-catenin、LRP表达均显著升高(P<0.05,P<0.01),第28日WIF1表达显著降低(P<0.05);与模型组比较,肺纤方中劑量组第14、28日大鼠肺组织wnt3a、β-catenin、LRP表达显著降低(P<0.05,P<0.01),WIF1表达显著升高(P<0.05,P<0.01)。结论 肺纤方具有抗肺纤维化的作用,其机制可能与抑制模型大鼠wnt3a、β-catenin、LRP的过表达和增加WIF1的低表达有关。

Abstract: Objective To investigate the effects of Feixian Formula on the expressions of wnt3a, β-catenin, WIF1 and LRP in bleomycin (BLM) -induced pulmonary fibrosis in rats; To explore its anti-pulmonary fibrosis mechanism. MethodsIntravenous intubation of BLM was used to establish pulmonary fibrosis rat models. 120 Wistar rats were randomly divided into sham-operation group, model group, Prednisone group, and Feixian Formula high-, medium- and low-dosage groups. The day after model establishing, gavage was intervened with corresponding medicine. Animals were treated in two batches on the 14th and 28th day after modeling. The protein expressions of wnt3a, β-catenin and WIF1 were detected by Western blot, and the mRNA expression of LRP was detected by RT-PCR. ResultsCompared with sham-operation group, the expressions of wnt3a, β-catenin and LRP in lung tissue of rats in model group increased significantly on day 14 and day 28 (P<0.05, P<0.01), and the expression of WIF1 decreased significantly on day 28 (P<0.05). Compared with model group, the expressions of wnt3a, β-catenin and LRP in lung tissue of rats in Feixian Formula medium-dosage group decreased significantly on day 14 and day 28 (P<0.05. P<0.01), and the expression of WIF1 increased significantly (P<0.05, P<0.01).
