核转录因子κB及其抑制因子研究(1)

     摘要:核转录因子κB(NFκB)是一类具有多向调节作用的转录调控因子,且在多个基因的转录及表达过程中发挥重要作用,与炎性相关疾病、肿瘤、疼痛等相关疾病相关。核转录因子抑制蛋白(IκB)主要由核转录因子κB抑制因子(NFκBIA)基因编码,参与调节NFκB信号通路,抑制NFκB功能,其同样与多种炎症性疾病、自身免疫性疾病及肿瘤的易感性有关。近年来,NFκB及其抑制因子在炎症、免疫及肿瘤等方面的影响一直广受关注。此外,NFκB信号通路与疼痛的调节也关系密切,但具体作用机制尚需进一步研究证实。本文主要对NFκB和IκB的结构、分布及表达情况,以及NFκB及NFκBIA基因在炎性疾病、肿瘤和慢性疼痛等多种疾病中的研究进展进行综述,旨在提高临床对其的认识。

    Nuclear Transcription Factor κB and Its Inhibitory Factors

    Abstract:Nuclear factor kappa-B (NFκB) is a multifaceted transcriptional regulator that plays an important role in the transcription and expression of multiple genes and is associated with inflammatory diseases, tumors, pain and other diseases.Nuclear transcription factor inhibitor protein (IκB), which is encoded by the Nuclear factor kappa-b inhibitor alpha (NFκBIA) gene, is involved in the regulation of the NFκB signaling pathway and the inhibition of NFκB function. It is also associated with a variety of inflammatory, autoimmune, and tumor susceptibility.The role of NFκB and its suppressor (NFκBIA) in inflammation, immunity, and tumor formation has received considerable attention in recent years.The NFκB signaling pathway is also closely related to pain regulation, but further studies are needed to confirm the specific mechanism.The structure, distribution and expression of NFκB and IκB were introduced and the research progress of NFκB and NFκBIA genes in inflammatory diseases, tumors and chronic pain are reviewed, aiming to improve clinical understanding of them.

    核轉录因子κB(nuclear factor κappa-B,NFκB)由NFκB基因编码,是一类具有多向调节作用的转录调控因子家族,在多个基因的转录及表达过程中发挥重要作用。1986年,NFκB首次被Sen R和Baltimore D从B细胞核中提取出来,因其能结合免疫球蛋白κ增强子的轻链基因,与细菌脂多糖(bacterial lipopolysaccharide,LPS)和放线菌酮联合作用导致超诱导,而被命名为核转录因子κB。近年来,NFκB作为最受关注的真核生物转录因子之一,被发现与免疫应答、炎症反应、细胞钙稳态、细胞分化和增殖、细胞凋亡、血管发生、肿瘤侵袭和转移等多个病理生理过程相关[1,2]。核转录因子抑制蛋白(IκB),主要由核转录因子κB抑制因子(nuclear factor kappa-B inhibitor alpha,NFκBIA)基因编码,参与调节NFκB信号通路,抑制NFκB功能。研究证实,IκB同样与多种炎症性疾病、自身免疫性疾病及肿瘤的易感性有关[1,3,4]。本文介绍NFκB和IκB的结构、分布及表达情况,并结合相关文献分析NFκB及NFκBIA基因与炎性疾病、肿瘤和慢性疼痛等多种疾病相关,可为临床诊断、治疗提供基因水平的辅助和治疗方向。, 百拇医药(石勤业 郭剑 徐建红)
