杜仲对UVA致HaCaT细胞光老化保护作用的实验研究(1)

     [摘要]目的:研究杜仲不同提取部位抗UVA致HaCaT细胞光老化的作用,并初步探讨其作用机制。方法:杜仲乙醇提取物经大孔树脂制备不同浓度乙醇梯度洗脱部位,分别作用于HaCaT细胞光老化模型,倒置显微镜下观察细胞形态,MTT法测定细胞活性,测定细胞培养液中LDH、MDA、SOD活力,确定并筛选杜仲抗皮肤光老化的作用及有效部位。结果:与模型组相比,浓度为1255μg/ml的50%乙醇大孔树脂洗脱部位能提高细胞活性(P<0.05),使培养液中LDH降低(P<0.01)、SOD活力提高(P<0.05)、MDA活力降低(P<0.05);浓度为125μg/ml的水洗脱部位使细胞培养液中LDH活力降低(P<0.05)。结论:杜仲抗皮肤光老化作用的有效成分主要集中在50%乙醇大孔树脂洗脱部位,其作用机制可能与其能清除氧自由基、保护细胞膜免受损伤有关。

    Protective effects of UVA induced photoaging in HaCaT cell by Eucommia ulmoides

    Abstract:ObjectiveTo research the resistance effects to ultraviolet radiation induced photoaging in HaCaT cell by different extract fractions from Eucommia ulmoides, and preliminarily discuss its functional mechanisms.MethodsEthanol extract of Eucommia macroporous resin were prepared by different parts of the concentration gradient of ethanol. Cell morphous was observed by inverted microscope; cell activeness was measured by MTT; values of LDH, MDA and SOD in culture solution were detected. And finally the anti-photoaging constitutes of Eucommia ulmoides were screened and confirmed.ResultsCompared to model group, 50% elution site (the concentration of 1255μg/ml) could enhance cell activeness (P<0.05), increase the value of SOD (P<0.05) and reduce the values of LDH (P<0.01) and MDA (P<0.05) in culture solution. 0% elution site (the concentration of 125μg/ml) could reduce the value of LDH (P<0.05) in culture solution.ConclusionsThe active constitutes of Eucommia ulmoides for skin anti-photoaging effects are mostly concentrated in 50% ethanol macroporous resin elution. The mechanisms are probably related to clearing oxygen free radicals and protecting cell membrane damages.

    皮肤光老化是指由于长期的日光照射导致皮肤加速衰老的现象,日光中的紫外线(UV,主要为UVA和UVB)是导致皮肤日晒伤、光老化及皮肤癌的重要因素。现代研究发现,杜仲具有促进胶原合成、抗衰老的作用,这些作用可能与杜仲的抗氧化活性有关[2]。本课题组前期工作表明,杜仲大孔树脂富集物能够抵抗UVB辐射引起的细胞氧化损伤,具有抗皮肤光老化的潜力 ......
