骨碎补总黄酮对骨质疏松大鼠Smad1 Smad5基因表达的影响(1)

     摘 要:目的:通过手术切除SD大鼠的双侧卵巢,建立绝经后骨质疏松模型,研究骨碎补总黄酮对骨质疏松大鼠Smad1、Smad5 mRNA表达的影响,从而在基因水平说明其影响成骨的机制。方法:(1)动物造模:60只12周龄SD大鼠,随机分成正常组、空白组和倍美力组及骨碎补总黄酮高、中、低剂量6组,每组10只,正常组麻醉后仅切除腹部与卵巢等重量的脂肪,其余50只麻醉后从腹部入路切除双侧卵巢。(2)处理方法:①正常组、空白组标准饲料常规饲养24周。②骨碎补总黄酮3组在用标准饲料喂养12周后,第13周开始在继续用标准饲料喂养的同时每天灌服骨碎补浓缩剂12周。③倍美力组在标准饲料常规喂养12周后,第13周开始在标准饲料常规喂养的同时每天灌服倍美力悬剂12周。④24周后处死大鼠切取右侧股骨,测Smad1、Smad5 mRNA。结果:空白组Smad1 mRNA的表达是正常组的40%,有统计学意义(P<0.05),Smad5 mRNA的表达是正常组的59.5%,有统计学意义(P<0.05);与空白组比较,骨碎补总黄酮高、中、低剂量组的Smad1、Smad5 mRNA表达均有提高,空白组与骨碎补总黄酮高剂量、中、低剂量组比较均有统计学意义(P<0.05),其中以骨碎补总黄酮中剂量组最为有效,分别为Smad1 mRNA(P<0.05)、Smad5 mRNA( P<0.01)。结论:骨碎补总黄酮能够上调骨髓微环境中Smad1、Smad5mRNA的表达水平,以中剂量组最明显,可能是其促进骨形成、修复骨损伤作用的机制之一。

    Effects of Drynariae Rhizoma Total

    Flavone on Smad1 Smad5 mRNA Expression in Osteoporotic Rats

    Abstract:Objective:Creating rat osteoporosis model by ovariectomy ,to study the effects of Drynariae Rhizoma Total Flavone on Smad1,5-mRNA expression in ovariectomied Rats, in order to explain its mechanism of bone generation in gene level.Methods:(1)Animal mode creating: Total of 60 SD rats 12 weeks old were divided to 6 groups randomly, 10 rats each: normal group, blank control group ,premarin group, high dose\\middle dose\\lower dose Drynariae Rhizoma Total Flavone group. All rats were ovariectomied except normal group.(2)Treating methods:① normal and blank control group were feed in general process for 24 weeks.②Drynariae Rhizoma Total Flavone group were feed with Drynariae Rhizoma Total Flavone in three kinds different concentration for 12 weeks after ovariectomied 12 weeks; ③Premarin group were feed with premarin for 12 weeks after ovariectomied 12 weeks.④ Kill rats and collect right femur bone to detect Smad1、Smad5. Results:Compared to the normal group ,Smad1 of blank control group is 40% (P<0.05), Smad5 of blank control group is 59.5% (P<0.05). Compared to the blank control group, All Smad1 and Smad5 expression in high dose\\middle dose\\lower dose Drynariae Rhizoma Total Flavone group are raised and have statistic meaning (P<0.05), significant increasing of Smad5 gene expression were documented in the middle dose Drynariae Rhizoma Total Flavone group(P<0.01).Conclusion:Drynariae Rhizoma Total Flavone seem to stimulate expression of Smad1 and Smad5 gene in bone marrow of ovariectomied rats, especially middle dose Drynariae Rhizoma Total Flavone, although the specific mechanism of promoting bone formation should be investigated further.

    骨质疏松症是严重影响老年人生活质量的常见病之一,它以骨量减少,骨组织显微结构退化,骨折危险度增加为特征,治疗非常棘手。中医药在治疗该病方面有明显优势。在众多补肾中药的研究中,不管是复方还是单味药,尽管疗效确切,但都存在药物成份的不明确性。因此,国内外科研工作者都试图研究一类不但疗效确切,而且成份明确的中药产品。骨碎补总黄酮就是这个背景下的产物,它是补肾中药骨碎补的提取物,有效成份明确,不含其它可能具有不同药理作用的成份,其提取物和提取工艺已获得国内国际专利,是国家九五科技攻关项目。, 百拇医药(朱慧锋 王维佳 王珠美)
