艾灸任脉经穴对脑缺血再灌注大鼠组织超微结构及巢蛋白表达的影响(1)

摘 要:目的:观察艾灸任脉经穴对脑缺血再灌注大鼠纹状体组织超微结构及侧脑室下区巢蛋白(Nestin)表达的影响。方法:采用线栓法建立大鼠大脑中动脉栓塞局灶性脑缺血再灌注模型。72只SD大鼠随机分为假手术组(Sham)、脑缺血再灌注模型组(MCAO)、模型+艾灸任脉治疗组(R)。用电镜观察脑缺血后组织超微结构及艾灸任脉对其的影响,并用免疫组化法观察艾灸任脉经穴对缺血侧侧脑室下区Nestin阳性表达的影响。结果:脑缺血损伤后各组大鼠纹状体的神经元、胶质细胞及血管等均可见不同程度的缺血损害,艾灸任脉经穴对于改善组织缺血损害具有一定的积极作用。模型组与假手术组相比,缺血侧侧脑室下区的Nestin的阳性表达均有不同程度的增加,其中再灌注7、14天差异有统计学意义(P<0.05);而艾灸任脉组的Nestin阳性表达于再灌注7、14、28天均明显多于模型组 (P<0.01)。结论:艾灸任脉经穴能够促进脑缺血大鼠侧脑室下区的神经干细胞的增殖,这可能是艾灸任脉经穴促进脑缺血损伤后神经修复的保护机制之一。

Abstract:Objective: To observe the effects of Ren vessel moxibustion on ultramicrostructure in corpora striata and nestin expression in subventricular zone (SVZ) after focal cerebral ischemia-reperfusion in rats. Methods:Cerebral ischemia-reperfusion injury model was induced by the method of nylon monofilament embolism. Seventy-two rats were divided randomly into sham operation group(Sham), ischemia-reperfusion model group(MCAO) and ischemia-reperfusion injury with treatment of Ren vessel Moxibustion group(R).We observed the ultramicrostructure of brain by electron microscope after focal cerebral ischemia-reperfusion and approached the effect of Ren vessel moxibustion on it. What's more, nestin positive expression in SVZ of the ischemic side was observed by immunohistochemistry. Results: After focal cerebral ischemia-reperfusion, the neuron, gliacyte and blood vessel in corpora striata all shew ischemic damage, and Ren vessel moxibustion had a positive effect to relieve ischemic damage ......
