摘 要:
目的:探讨穿心莲内酯联合5-Fu对人低分化胃癌细胞株BGC-823的协同抗肿瘤作用。 方法:以人胃癌BGC-823细胞株为靶细胞,通过体外细胞培养和裸鼠移植瘤试验,采用 MTT法检测、绘制效应-合用指数曲线、抑瘤率计算等方法。 结果:穿心莲内酯在7.5～120μg/mL、5-FU在3.75～60μg/mL范围内均能抑制BGC-823细胞的生长,具有剂量-效应关系和时间-效应关系。两药联合作用24h、48h、72h时的中效抑制浓度分别为17.32、9.54、4.11μg/mL,显著小于单药应用组。72h效应-合用指数曲线表明两药在抑制效应≤0.97范围内可产生协同作用。穿心莲内酯联合5-FU对人BGC-823细胞裸鼠移植瘤的抑瘤率为(75.51±12.57)%,与单药组相比差异均有显著性。结论:穿心莲内酯、5-Fu能在体内外抑制人胃癌BGC-823细胞的生长,两药联合产生协同或相加作用。

Synergistic Effect of Andrographolide and 5-fluorouracil on the
Inhibition of BGC-823 Cell line of Human Gastric Cancer

Abstract:Objective:To study the anti-proliferation effects of the Andrographolide with the 5-fluorouracil(5-Fu) on BGC-823 cell line of human gastric carcinoma. Methods:MTT assay were applied to determine cell inhibition in vitro. Median inhibitory concentrations (IC50 ) of andrographolide with 5-FU or used alone were calculated; Tumor inhibition rate of xenografts in nude mice were measured in vivo. Results:Andrographolide at the dose of 7.5～120μg/mL and 5-Fu at the dose of 3.75～60μg/mL can both inhibit the proliferation of BGC-823 cells with a concentration and time dependent manner. The cell inhibition rates and xenografts inhibition rates (75.51±12.57)% was obviously increased when the two agents were combined used. Median inhibitory concentration (ic50)of andrographolide with 5-FU at the time of 24h, 48h, 72h were17.32,9.54,4.11μg/mL respectively, which were significantly lower than single groups. The curve of combination index-effect relationship indicated a synergistic effect produce by andrographolide and 5-FU when the response rate was lower than 97% ......
