壮骨镇痛胶囊对乳腺癌细胞迁移运动信号通路关键蛋白p-JNK和Rac-1表达的影响(1)

摘 要:目的:研究壮骨镇痛胶囊对高转移人乳腺癌MDA-MB-231细胞迁移及其运动信号通路关键蛋白p-JNK和Rac-1表达水平的影响。方法:采用划痕法测定高(5.0%)、中(1.25%)、低(0.63%)不同浓度壮骨镇痛胶囊含药血浆对MDA-MB-231细胞迁移的影响,采用免疫细胞化学法检测不同浓度壮骨镇痛胶囊对MDA-MB-231细胞p-JNK和Rac-1蛋白表达水平的影响。结果:高、中浓度壮骨镇痛胶囊含药血浆显著抑制了乳腺癌MDA-MB-231细胞的迁移和p-JNK、Rac-1蛋白表达,低浓度血浆对细胞迁移和p-JNK、Rac-1蛋白表达无显著影响。结论:壮骨镇痛胶囊可通过改变运动信号通路关键蛋白p-JNK和Rac-1的表达来抑制乳腺癌MDA-MB-231细胞的迁移,且该作用与浓度密切相关。

The Effects of Zhuanggu Zhentong Capsules on the Expressions of Key Proteins in Migrating-Related Signaling Pathway Such as p-JNK and Rac-1of Breast Cancer Cell Lines in Vitro

Abstract:Objective:To study the impacts of Zhuanggu Zhentong Capsules(ZZC) on the cell migration and the expressions of key proteins in migrating-related signaling pathway such as p-JNK and Rac-1 of breast cancer cell lines MDA - MB -231 in vitro. Methods:The impacts of different doses of ZZC contained plasma (5.0%,1.25% and 0.63%) on the migrations of MDA - MB -231 cells in intro were measured by wound-healing assay. These proteins levels were assayed by immunocytochemistry protocol. Results: The cell migration and the expressive levels of p-JNK and Rac-1proteins in MDA - MB -231 cells treated with 5% ZZC contained plasma and 1.25% ZZC contained plasma were inhibited significantly. However, there are no obvious inhibitions on the cell migrations and p-JNK and Rac-1 proteins expressions in MDA - MB -231 cells treated with 0.63% ZZC contained plasma. Conclusion: ZZC contained plasma can inhibit the migrations of MDA - MB -231 cells in dose - dependent manner by changing the expressions of key proteins in migrating-related signaling pathway such as p-JNK and Rac-1 proteins.
