丹星通络汤治疗大鼠脑缺血再灌注Bax蛋白表达的影响(1)

摘要:目的 观察丹星通络汤对大鼠脑缺血再灌注损伤海马区细胞凋亡调控基因Bax蛋白表达的影响，探讨丹星通络汤预防与治疗大鼠脑缺血再灌注损伤的作用。方法 SD大鼠40只，随机分成假手术组、模型组、尼莫地平组、丹星通络汤小剂量组、丹星通络汤大剂量组，每组8只。采用线栓法制备大鼠大脑中动脉闭塞(MCAO)再灌注模型。HE染色检测大鼠脑组织海马区的病理学变化，免疫组织化学法和医学图像分析法检测大鼠脑组织海马区Bax蛋白的平均光密度(OD)值。结果 与模型组比较，丹星通络汤大、小剂量组大鼠脑组织海马区Bax蛋白的OD值明显减低(P<0.05)，并且丹星通络汤大剂量组与尼莫地平组相比有统计学意义(P<0.05)。结论 丹星通络汤可能通过下调Bax蛋白的表达，从而抑制脑组织的凋亡机制发挥对脑组织的保护作用。

The Influence of Danxing Tongluo Decoction on the Protein Expression of Bax during Cerebral Ischemic Reperfusion Injury in Rats

Abstract：Objective To observe effect of Danxing Tongluo decoction(DTD)on the expression of Bax and the neuronal apoptosis during cerebral ischemic reperfusion injury in rats.Methods Forty SpragueDawley (SD) rats were randomly divided into shamoperation group,model group,nimodipine group,DTD low dosage group,and DTD high dosage group.The cerebral ischemia was induced by right middle cerebral artery occlusion for 2 hours and reperfusion 24 hours in rats except shamoperation group.Pathological changes in rat hippocampus were observed.The mean optical density (OD) of Bax protein in rat hippocampus was measured by immunohistochemistry and Image Pro Plus (IPP).Results The OD value of Bax proteins in brain tissue of rats in treatment groups were reduced compared with that in model group (P<0.05),and that in nimodipine group was lower than that in DTD high dosage group (P<0.05).Conclusion DTD can inhibit the neuronal apoptosis by inhibiting the expression of Bax protein in cerebral ischemic reperfusion injury.And this is one of protective mechanism of DTD against cerebral ischemic reperfusion injury.

缺血性脑血管病属于中医的“中风”范畴，中风病理性质多属本虚标实。肝肾阴虚，气血衰少为致病之本，风、火、痰、气、瘀为发病之标，两者可互为因果。导致阴阳失调、气血逆乱，出现昏仆、半身不遂、口眼歪斜等症。其中血瘀贯穿病程始终，所以治疗多采用活血化瘀法。丹星通络汤具有通腑化瘀、祛痰开窍的作用，使腑气通、痰热去、九窍利。在临床上对预防和治疗缺血性脑血管病具有一定疗效，但作用机制尚未阐明。Oltvai等[1]发现Bax的高表达可加速细胞的凋亡。本文通过观察丹星通络汤对脑缺血再灌注损伤海马区Bax蛋白表达的影响，进一步探讨丹星通络汤保护脑组织作用的机制。
