柱前衍生反相高效液相色谱法测定血浆谷胱甘肽
[摘 要] 目的:建立一种柱前衍生反相高效液相色谱法测定血浆同型谷胱甘肽(glutath ione,GSH)的方法。方法:以tris-(2-carboxy lethy l)-phosphine(TCEP)为还原剂,7-fluorbenzo-2-oxa-1,3-diazole-4-sulfonate(SBD-F)为衍生剂,N-乙酰半胱氨酸(N-ace tylcystine,NAC)为内标,C8柱分离。流动相为甲醇-磷酸盐缓冲液,梯度洗脱,激发波长(λex)380nm,发射波长(λem)510nm,内标标准曲线法定量。结论:本方法准确、灵敏、快速、特异,适合于GSH实验室研究和临床常规检测。[关键词] 柱前衍生反相高效液相色谱法;谷胱甘肽
[Abstract] Objective Establish a pre-column derivatization RP-HPLC with plasma glutathione (glutath ione, GSH) method.Method With tris-(2-carboxy lethy l)-phosphine (TCEP) as reductant,7-fluorbenzo-2-oxa-1 ,3-diazole-4-sulfonate (SBD-F) as a derivative agent, N-acetyl cysteine acid (N-ace tylcystine, NAC) as an internal standard, C8 column separation. Mobile phase was methanol - phosphate buffer, gradient elution, the excitation wavelength (λex) 380nm, emission wavelength (λem) 510nm, internal standard quantification standard curve. Conclusion This method is accurate, sensitive, rapid, specific, suitable for laboratory research and clinical practice GSH detection. ......
