摘要:目的以栉杆菌多糖(APD)为研究对象,进行硫酸酯化(SAPD),并测定其抗肿瘤活性和免疫调节活性。方法APD采用氯磺酸-吡啶法进行酯化,溴化二苯偶氮盐(MTT)比色法研究样品在体外对肿瘤细胞株的抑制作用和促进正常小鼠脾脏细胞增生的作用。结果红外光谱证明SAPD已经形成硫酸酯,其取代度为0.918。APD对肿瘤细胞株几乎无细胞毒性,SAPD对A549肺癌细胞株的抑制率为31%。APD对ConA 诱导的脾脏细胞增生具有促进作用,但对LPS诱导的脾脏细胞增生无促进作用。SAPD低浓度时对ConA诱导的脾脏细胞增生有促进作用,对LPS诱导的脾脏细胞增生无促进作用。结论APD经过硫酸酯化后抗肿瘤活性显著增强,对ConA诱导的脾脏细胞增生有一定的促进作用。

Sulfation of Polysaccharide from Arthrobacter sp. and Comparative Study on Bioactivity of Polysaccharide and Its Sulfated Derivative

Abstract:Objective To study the sulfation of the polysaccharide from Arthrobacter sp.(APD) and determine the antitumor and immunity modulation activities of it and its sulfated derivative (SAPD). MethodsAPD was chemically modified by chlorosulfonic acid-pyridine method. MTT colourimetry was used to evaluate the antitumor activity against human tumor cells in vitro,as well as the proliferation-enhancing effect on spleen lymphocytes of normal mice. Results The IR spectrum showed that SAPD had been formed, the substituted degree was 0.918. APD had little cytotoxic activity towards the cancer cells, while SAPD inhibited the growth of A549 lung cancer cells remarkably with 31% inhibitory ratio. Though APD had no effect on the proliferation of LPS-induced spleen cells, it could enhance the proliferation of ConA-induced spleen cells. SAPD with low dose could promote the proliferation of ConA-induced spleen cells, but also had no effect on the proliferation of LPS-induced spleen cells. ConclusionThe antitumor activity of APD is remarkably enhanced after it was sulfated. APD can also promote the proliferation of ConA-induced spleen cells.

多糖的硫酸酯化是多糖化学修饰研究中的一个热点,实验表明硫酸酯化多糖具有抗病毒、抗肿瘤、抗凝血等作用。本实验对栉杆菌中的多糖进行分离纯化,并对其中一种组分(APD)进行硫酸酯化,得到硫酸酯化多糖(SAPD),然后对APD和SAPD的抗肿瘤活性和免疫活性进行了初步研究。
