摘要:目的研究葡萄籽提取物的抗氧化功能。方法以SPF级雄性昆明种小鼠为实验动物,根据小鼠体重随机分为5组,每组10只,分别是空白组、模型组和低、中、高3个剂量组,空白组和模型组每天经口给予溶剂植物油,剂量组每天经口给予葡萄籽提取物,连续灌胃30 d,取血测抗氧化酶活性。此后除空白对照外,各组射线全身一次性照射5~8 Gy 60Coγ,照射后第4天处死动物,取肝组织测过氧脂质含量和抗氧化酶活性,比较各剂量组与模型组的差别。结果经口给予小鼠葡萄籽提取物30 d,高、中剂量组小鼠血中超氧化物歧化酶(SOD)活性,以及高剂量组小鼠血中谷胱甘肽过氧化物酶(GSH-Px)和肝组织中GSH-Px、SOD活性均明显高于模型组,各剂量组小鼠肝中丙二醛(MDA)水平均明显低于模型组。结论葡萄籽提取物有降脂质过氧化的作用,有抗氧化功能。

Empirical Study on Anti-oxidative Effect of Grape Seed Extract

Abstract:Objective To study the anti-oxidative effect of grape seed extract. Methods Fifty mice were randomly divided into five groups of normal control , model control and low, moderate, high dose groups. In normal control and model control groups, corn oil was given orally. In low, moderate, high dose groups, grape seed extract was given orally. After 30 days, the activities of GSH-Px , SOD in blood were measured. Then, all the groups except normal control group received one-time 5~8Gy60C0 γ ray for the whole bodies. At the fourth day after radiation, all the mice were killed and the activities of GSH-Px, SOD in hepatic tissues were measured. The difference among all groups was compared. Results After 30 days, SOD activity in blood in moderate and high dose groups, and the activities of GSH-Px in blood and GSH-Px, SOD in hepatic tissues in high dose group were higher than that in model control group. The level of malonaldehyde (MDA) in hepatic tissues in all dose groups was lower than that in model control group. Conclusion Grape seed extract can inhibit the lipid peroxidation and have an anti-oxidative effect.

现代医学证明,多种疾病的发生发展与氧化损伤有关[1]。适当补充抗氧化剂对提高机体抗氧化能力,增进身体健康有重要意义。20世纪50年代以来,人们从葡萄果实、叶和其它部位分离、鉴定出的成分达100多种,发现有些成分具有生物活性,显示出抗氧化、抗高脂血症、抗疲劳和抗肿瘤等多种作用[2]。其中葡多酚(GPC),国外多称原花青素,是存在于葡萄中的天然植物多酚,在葡萄籽中含量最丰富,是葡萄籽提取物的主要成分。国外报道,葡多酚在体外具有清除自由基及抗脂质过氧化[3]等活性。研究表明[4],原花青素还可以增强超氧化物歧化酶(SOD)、谷胱甘肽-S-转移酶等的活性 ......
