樱草杜鹃黄酮类成分的提取工艺研究(1)

[摘要] 目的:优选樱草杜鹃中总黄酮的最佳提取工艺。方法:以芦丁为评价指标,考察黄酮质量分数,通过正交试验对超声法提取黄酮的工艺进行优化。结果:超声提取法的最佳工艺条件为以樱草杜鹃叶为原料,75%乙醇为提取液,m(樱草杜鹃)∶v(提取液)=1∶10,超声时间为60 min,提取3次;在该条件下,总黄酮的提取率为11.01%,RSD为0.43%(n=3)。结论:优选了樱草杜鹃总黄酮的最佳提取工艺,提高药材的利用率,为更好地利用该药材提供了理论依据。

Optimal technology for extracting flavonoids from Rhodendron primulaeflorum Bur. et Franch.

[Abstract] Objective: To optimize the best extraction technology of flavonoids in leaves of Rhodendron primulaeflorum Bur. et Franch. Methods: For the evaluation of rutin to study the mass fraction of flavonoids, ultrasonic extraction process was optimized flavonoids by orthogonal test. Results: The optimum ultrasonic extraction conditions as follows: the Rhodendron primulaeflorum Bur. et Franch. leaves was as raw material, 75% ethanol for the extract, m (Rhodendron primulaeflorum Bur. et Franch.)∶v (extract) was 1∶10, ultrasonic was 60 min, extracted three times; in this condition, the extraction rate of flavonoids was 11.01% with a RSD of 0.43%(n=3). Conclusion: Optimize the best extraction technology of flavonoids in Rhodendron primulaeflorum Bur. et Franch. leaves and improve the utilization of medicinal herbs in order to better use of the medicine provides a theoretical basis.

“达里”始载于八世纪的《月王药珍》[1],现代藏医药著作《甘露本草明镜》[2]称其“花味甘、苦、涩,消化后变为甘,功效轻、热、干,主治浮肿、水土不适、肺病、气管炎、体弱、声哑。叶味苦、涩,消化后变苦,功效热、锐,治胃寒、饮食不正、皮肤病、四肢僵硬”。樱草杜鹃(Rhodendron primulaeflorum Bur. et Franch.)为杜鹃花科杜鹃属植物,著名藏药“达里”的原植物之一,分布于我国云南、四川、西藏[3]。黄酮类化合物为药材樱草杜鹃的主要药效成分,其中槲皮素为黄酮类化合物的主要代表性物质[4]。本课题组也从藏产樱草杜鹃中分得槲皮素等多种黄酮类化合物且含量较大。目前尚未见对其黄酮类化合物的提取工艺研究的报道,本文以芦丁为评价依据,以黄酮质量分数为指标,利用正交试验对樱草杜鹃中总黄酮提取的影响因素进行了考察,确定了超声提取樱草杜鹃中总黄酮最佳提取工艺,为更好地利用该药材提供了理论依据 ......
