[摘要] 目的:探讨TIE2受体蛋白在野百合碱诱导的肺动脉高压中的作用。方法:应用野百合碱诱导的慢性肺动脉高压大鼠模型,免疫组化观察肺动脉构型重建及细胞凋亡,应用RT-PCR技术检测大鼠肺动脉TIE2 mRNA表达。结果:野百合碱大鼠肺动脉压明显升高,免疫组化观察到肺血管重构及内皮细胞层Caspase-3蛋白表达,野百合碱诱导的肺动脉高压大鼠肺血管TIE2 mRNA表达明显减少。结论:在野百合碱诱导的肺动脉高压大鼠模型中,肺血管重构,内皮细胞损伤凋亡可能与肺血管TIE2受体表达降低有关。

Expression of TIE2 gene in monocrotaline-induced pulmonary hypertension

[Abstract] Objective: To investigate the effect of TIE2 gene in monocrotaline (MCT)-induced pulmonary hypertension. Methods: MCT-treated rats were used as a model for chronic PHT. The remodeling and cell apoptosis of pulmonary arteries (PAs) were analyzed by immunohistochemical method. RT-PCR was performed to identify the expression of TIE2 mRNA in rat PAs. Results: The pulmonary hypertension was established in MCT rats. The remodeling and protein expression of Caspase-3 were observed in PAs. A lower level of TIE2 mRNA expression was detected in PAs from MCT rats. Conclusion: The lower level of TIE2 mRNA expression might contribute to the remodeling and endothelial cell apoptosis of PAs in MCT-induced pulmonary hypertension.

肺动脉高压(pulmonary hypertension,PHT)是一种进行性、消耗性疾病,表现为肺动脉血管阻力持续升高,最终导致右心衰竭,直至死亡[1]。肺动脉平滑肌细胞(PASMC)增生及血管收缩增强引起血管构型重建的过程是形成PHT的主要原因[2]。TIE(tyrosine kinase with Ig and EGF homology domains)是血管内皮细胞上特异的酪氨酸激酶型受体,是Ang家族的受体,广泛表达于各级血管内皮细胞中[3]。TIE受体被活化后,激活胞内的第二信使系统,具体说就是激活了内皮细胞中丝裂原激活蛋白激酶(mitogen activated protein kinases,MAPK)的至少3种亚型,它们分别在与应力相关的作用中、细胞骨架的重建、细胞间的黏附与迁移、内皮细胞增殖中发挥作用[4]。本实验通过观察野百合碱(monocrotaline,MCT)诱导的PHT大鼠TIE受体表达的变化,探讨TIE2受体在PHT发生及发展中可能的作用和机制。
1 材料与方法
1.1 建立PHT大鼠模型
雄性SD大鼠20只(大连大学实验动物中心提供),体重(150±10) g,随机分为两组。实验组一次性腹腔内注射MCT 60 mg/kg(Wako Pure Chemical Research Company,JAP),对照组注射同等剂量的溶剂。大鼠常规给予食物和水,保持温度(18~22℃)和湿度(50%~70%),饲养3周[5]。
1.2 MCT大鼠模型肺动脉平均压和右心指数检测
