[摘要] 目的:建立高效液相法测定小儿布洛芬口服溶液中布洛芬含量的方法。方法:采用CenturySIL ODS2(4.6 mm×200 mm,5 μm)色谱柱;流动相:醋酸钠缓冲液(取醋酸钠6.13 g,加水750 ml,振摇使溶解,用冰醋酸调节pH值至2.5)-乙腈(40∶60);检测波长:265 nm;流速:1.0 ml/min;柱温:30℃。结果:布洛芬在0.2~1.4 mg/ml范围内与峰面积呈良好的线性关系(r=0.999 9),平均回收率为98.1%,RSD=1.2%(n=9)。结论:本法简便、准确、重现性好,可用于小儿布洛芬口服溶液中布洛芬的质量控制。

Content determination of Ibuprofen in Paediatric Ibuprofen Oral Solution by HPLC

[Abstract] Objective: To establish a method for the content determination of Ibuprofen in Paediatric Ibuprofen Oral Solution by HPLC. Methods: The CenturySIL ODS2 (4.6 mm×200 mm, 5 μm) column was used. The mobile phases consisted of sodium acetate buffer solutions (Weighed sodium acetate 6.13 g , added water 750 ml, shake for dissolution, adjusted with glacial acetic acid to a pH of 2.5) and acetonitrile(40∶60), the flow rate was 1.0 ml/min and the detection wavelength was at 265 nm, the column temperature was 30℃. Results: The linear ranges of Ibuprofen was at 0.2~1.4 mg/ml (r=0.999 9), the average recovery was 98.1% with a RSD of 1.2%(n=9). Conclusion: The method is fast, reliable, accurate and can be used in the quality control of Ibuprofen in Paediatric Ibuprofen Oral Solution.

布洛芬(Ibuprofen,简称IB),是一种非甾体抗炎药。其作用机制主要通过对环氧酶的抑制而减少前列腺素的合成,从而降低神经痛觉的敏感性,同时通过下丘脑体温调节而起解热作用。小儿布洛芬口服溶液为长葛市人民医院制剂,由布洛芬、氢溴酸右美沙芬、扑尔敏、醋酸钠、蜂蜜、白糖、注射用水组成,具有镇痛、抗感染、解热等作用,用于小儿感冒引起的发热、头疼、咳嗽、鼻塞、流鼻涕、流眼泪、打喷嚏等症状。疗效确切,给药剂量容易控制,特别适合儿科使用,为了控制该制剂质量,保证临床用药安全有效,故选择该制剂中的主要成分布洛芬作为指标成分,进行含量分析研究。本文建立了小儿布洛芬口服溶液中布洛芬含量测定的方法。结果表明,该法专属性强,干扰小,并具有较好的重复性,可用于小儿布洛芬口服溶液的质量控制。
1 仪器与试药
1.1 仪器
UVIKON-XL紫外-可见分光光度计;日本岛津LC-10A液相色谱仪(LC-10ATvp泵,SPD-10Avp紫外-可见检测器,CTO-10ASvp柱温箱,Chromato-Solution Light 色谱工作站);BP211D电子天平。
1.2试药
布洛芬对照品(中国药品生物制品检定所,批号:0179-9702);小儿布洛芬口服溶液(长葛市人民医院制剂室);缺布洛芬的阴性样品(长葛市人民医院制剂室);乙腈为色谱纯,水为超纯水,其余试剂均为分析纯。
