[摘要] 目的:建立复方逍遥合剂中黄芩苷的含量测定方法。方法:采用HPLC法,色谱柱Calesil-C18(4.6 mm×250 mm,5 μm),流动相:甲醇∶0.1%磷酸(47∶53),检测波长:280 nm。结果:黄芩苷进样量在0.08~0.80 μg范围内呈良好的线性关系(r=0.999 9 ),低、中、高三种浓度的平均回收率为99.59%、100.22%、100.40%,RSD分别为0.75%、2.58%、0.86%。结论:本法简便、准确,重复性好,可用于该产品的质量控制。

Content determination of Baicalin for Compound Xiaoyao Oral Solution by HPLC

[Abstract] Objective: To establish a method for determination the content of Baicalin in Compound Xiaoyao Oral Solution.Methods: Calesil-C18 column (4.6 mm×250 mm,5 μm) was used,Mobile phase was methanol-0.1% phosphoric acid (47∶53),Detection wavelength was 280 nm. Results: Baicalin sample size located 0.08-0.80 μg,and favourable linear correlation (r=0.999 9) was showed. The average recovery rates of low, moderate and high concentration were 99.59%, 100.22%,100.40% with RSD of 0.75%, 2.58%, 0.86%, respectively. Conclusion: The method is convenient and accurate. Meanwhile, it also has good reproducibility, may be used to quality control of Baicalin.

复方逍遥合剂为我院自制制剂(沪药制字Z04090765),由黄芩、当归、郁金、陈皮、香附、柴胡、白芍、柴胡、栀子、茯苓、生牡蛎 、黄柏、白术等十几味中药组成。具有舒肝理气、清热活血、软坚化痰等功能,用于调理肝郁痰阻型女童早期性早熟,运用临床多年来效果显著。其中黄芩为处方主药,黄芩苷是黄芩主要有效成分。为控制产品质量,确保临床疗效,参考文献[1-3],本文采用HPLC法对制剂中黄芩苷含量进行定量分析,方法简便、准确、重复性好,可用于该产品的质量控制。
1 仪器与试药
1.1 仪器
Agilent 1100高效液相色谱仪,包括Agilent G1311A 四元泵(Quatpump),Agilent G1322A 真空在线脱气机(Degrasser),Agilent G1313A 自动进样器(ALS),Agilent G1315B DAD二级管阵列检测器,Agilent chemstation system(安捷伦化学色谱工作站);电子天平(Sartorius,d=0.01 mg);超声提取器(SB1000,宁波新芝生物科技股份有限公司)。
1.2 试药
供试品复方逍遥合剂 (上海市儿童医院);黄芩苷对照品(中国药品生物制品检验所,批号:110715-200815);甲醇为色谱纯,水为重蒸水,其他试剂均为分析纯。
2 方法与结果
2.1 色谱条件[4]
色谱柱Calesil-C18(4.6 mm×250 mm,5 μm); 流动相:甲醇-0.1%磷酸(47∶53);检测波长:280 nm;柱温:30℃;流速:1 ml/min;理论板数按黄芩苷峰计算不低于2 000。
2.2 溶液制备[5-6]
2.2.1 对照品溶液制备精密称取在五氧化二磷真空干燥24 h的黄芩苷对照品0.004 00 g,加甲醇8 ml,超声20 min ......
