缺血对胰岛素抵抗大鼠Apelin mRNA、APJ mRNA表达的影响(1)

     [摘要] 目的:研究胰岛素抵抗(insulin resistance,IR)大鼠心肌Apelin、孤儿G蛋白偶联受体(APJ)表达及缺血对它的影响。方法:将24只SD大鼠随机分三组,即IR组、IR+缺血组和对照组,每组8只。采用Real-time-PCR法测定心肌组织Apelin mRNA和APJ mRNA表达水平。结果:IR组Apelin mRNA、APJ mRNA相对表达量较对照组升高,两组比较,差异有统计学意义(P<0.05)。IR+缺血组Apelin mRNA、APJ mRNA相对表达量较对照组降低,两组比较,差异有统计学意义(P<0.05)。结论:大鼠心肌组织存在Apelin、APJ表达。在胰岛素抵抗大鼠中,心肌组织Apelin、APJ代偿性表达增加,推测Apelin、APJ可能参与了胰岛素抵抗的发生,对心脏发挥保护性作用。胰岛素抵抗大鼠心肌缺血后,心肌组织Apelin、APJ表达明显下降,推测可能与Apelin代偿不足或由于肾素-血管紧张素-醛固酮系统激活,反馈性抑制Apelin、APJ表达有关,Apelin、APJ对心脏的保护作用减弱。

    Effects of ischemia on the expression of Apelin mRNA and APJ mRNA in insulin-resistant rats

    [Abstract] Objective: To study the expression of Apelin and APJ in the myocardial tissue of insulin-resistant rats and the effects of ischemia on it. Methods: 24 SD rats were randomly divided into three groups, IR group, IR+ischemia group and control group. The expression of APJ mRNA and Apelin mRNA were tested with Real-time PCR method. Results: Compared with the control group, Apelin mRNA relative expression, APJ mRNA relative expression in insulin resistance group increased, there was a significant difference (P<0.05). Compared with the control group, Apelin mRNA relative expression, APJ mRNA relative expression in IR+ischemic group significantly decreased, there was a significant difference (P<0.05). Conclusion: There are the expression of Apelin and APJ in the myocardial tissue of rats. The expression of Apelin and APJ was increased in insulin resistance rats. Apelin and APJ may be involved in the occurrence of insulin resistance to protect the heart. Myocardial Apelin and APJ expression significantly decrease in insulin-resistant rats after myocardial ischemia, suggest that Apelin may be inadequate or the renin-angiotensin-aldosterone system is activated to inhibit the expression of Apelin and APJ, which weaken the protective effect of Apelin and APJ on the heart ......
