1型糖尿病小鼠骨髓EPCs的培养和功能研究(1)
[摘要] 目的 研究1型糖尿病小鼠骨髓内皮祖细胞(EPCs)的培养方法和血管原性功能。 方法 采用7～8周雄性C57/B6小鼠，腹腔注射链脲佐菌素建立1型糖尿病小鼠模型，同时设立正常对照组，连续5 d给等量的柠檬酸缓冲液。采用密度梯度离心法分离并培养糖尿病小鼠骨髓EPCs；通过MTT法、Matrigel、改良Boyden小室分别检测EPCs的增殖、迁移和小管形成的功能性指标。 结果 与正常对照组比较，培养获得1型糖尿病小鼠的EPCs增殖减少，其迁移和小管形成能力明显下降(P < 0.05)。 结论 成功培养1型糖尿病小鼠骨髓EPCs，EPCs的血管原功能受损。
