牙龈卟啉单胞菌超声提取物对人牙周膜细胞活性的影响

     [摘要]目的探讨牙龈卟啉单胞菌超声提取物对人牙周膜细胞活性的影响。方法组织块法体外培养人牙周膜细胞(hPDLC),以6.25、12.5、25、50 mg·L-1质量浓度的超声提取物作用于hPDLC 24 h,阴性对照组为无超声提取物作用的hPDLC;用12.5 mg·L-1质量浓度的超声提取物分别作用于hPDLC 24、48、72、96 h,阴性对照组亦培养24、48、72、96 h。以甲噻唑四唑氮比色比较试验组与对照组hPDLC的活性变化,用SPSS 13.0软件包对数据行单因素方差分析和独立样本t检验。结果当作用于hPDLC的超声提取物质量浓度为6.25~50mg·L-1时,经过24 h,试验组hPDLC的活性低于阴性对照组;当超声提取物质量浓度为12.5 mg·L-1时,在24~96 h内,试验组hPDLC的活性低于阴性对照组。结论牙龈卟啉单胞菌可导致体外培养的hPDLC的活性降低,且作用呈质量浓度和时间依赖性。

    [Abstract]ObjectiveTo evaluate the effects of sonicated extracts of Porphyromonas gingivalis(P.gingivalis)on the viabilities of human periodontal ligament cells(hPDLC). MethodsIn the experimental group, hPDLC obtained by the tissue culture were treated by sonicated extracts in different concentrations(6.25, 12.5, 25, 50 mg·L-1)for 24 hours. In the negative control group, hPDLC were treated without sonicated extracts. Furthermore, hPDLC were exposed to 12.5 mg·L-1sonicated extracts for 24, 48, 72 and 96 h, while hPDLC in the negative group were cul -tured for 24, 48, 72 and 96 h without sonicated extracts. Cell viabilities in two groups were determined by methyl thiazolyl tetrazolium method and use SPSS 13.0 package for one-way analysis of variance and independent sample t-test. ResultsSonicated extracts for hPDLC as the concentration of 6.25-50 mg·L-1, exposed for 24h, the hPDLC viabilities of experimental group were lower than those of the negative group. When hPDLC were exposed to 12.5 mg·L-1sonicated extracts, in the 24-96 h, the hPDLC viabilities of experimental group were lower than those of the control group.
