摘要目的:观察纳米雄黄对乳腺癌MCF-7细胞上皮间质转化的影响,并探讨其相关机制。方法:体外培养乳腺癌MCF-7细胞,不同浓度纳米雄黄溶液干预,并设立对照组,免疫组化实验检测各组细胞E-cad、HIF-1α表达情况,RT-PCR检测SnailmRNA表达情况。结果:各纳米雄黄组E-cad表达阳性率均明显高于对照组(P<0. 01),而HIF-1α表达阳性率均低于对照组(P<0. 01);对照组、纳米雄黄低、中、高剂量组、阿霉素组的SnailmRNA相对表达量依次为(52. 16±3. 87)、(30. 10±2. 48)、(47. 08±3. 45)、(41. 42±4. 35)、(36. 07±2. 45)。结论:纳米雄黄可通过上调E-cad表达、下调HIF-1α 及Snail mRNA表达逆转乳腺癌MCF-7细胞上皮间质转化发挥抗肿瘤转移作用。
