益气活血药物对脑梗死恢复期大鼠cfos基因表达的影响(1)
摘要目的:基于cfos mRNA觀察益气活血药物对脑梗死恢复期大鼠的影响。方法:以Longa的方法略作改良制作大鼠左侧大脑中动脉阻塞模型,按照随机数字表的方法随机分为正常组、模型组、假手术组、益气组、活血组、益气活血组、ERK抑制剂7组,采用RTPCR技术,在治疗28 d观察益气活血药物对脑梗死恢复期大鼠梗死皮层cfos mRNA表达的影响。结果:在治疗28 d后,模型组较正常组cfos mRNA表达明显下降(P<005),益气活血组与ERK抑制组较模型组有一定程度提高(P<005),活血组与模型组比较,无明显统计学意义(P>005),益气组的cfos mRNA表达低于其他各组(P<005)。结论:在脑梗死恢复期,益气活血药物在一定程度上可改善脑缺血,优于单纯益气与单纯活血,其疗效机制可能与提高cfos mRNA的表达有关。

AbstractObjective:To observe the expression of cfos mRNA of rats with cerebral infarction in recovery period under supplementing qi and activating blood circulation drugs. Methods:Longa method was slightly modified in making the rat model of left middle cerebral artery occlusion. According to the random number table method,the rats were randomly divided into 7 groups,including a normal group, a model group, a sham operation group, a supplementing qi group, an activating blood circulation group, a supplementing qi and activating blood circulation group, and an ERK inhibitor group. Using RTPCR technique, we observed the effect of supplementing qi and activating blood circulation drugs on cfos mRNA of rats with cerebral infarction after 28 days′ treatment in myocardial infarction cortex. Results:After 28 days′ treatment,compared with normal group, cfos mRNA expression in the model group was significantly decreased (P<005);cfos mRNA expression in the supplementing qi and activating blood circulation group and ERK inhibition group increased to a certain extent than that in the model group (P<005);the activating blood circulation group had no significant statistical difference with the model group(P>005);cfos mRNA expression in supplementing Qi group was lower than the other groups (P<005). Conclusion:In the recovery period of cerebral infarction, supplementing qi and activating blood circulation drugs can improve cerebral ischemia in a certain degree, which is better than simply supplementing qi and activating blood circulation, and its therapeutic mechanism may be related to improving the expression of cfos mRNA.
