依达拉奉对脑出血大鼠血肿周围组织NF-κB p65蛋白表达的影响

    [摘要] 目的 探讨依达拉奉对脑出血大鼠血肿周围组织NF-κB p65蛋白表达的影响。 方法 108只健康雄性Wistar大鼠随机分为正常组、假手术组、脑出血组和依达拉奉治疗组,其中脑出血组和依达拉奉治疗组又分为3 h、6 h、12 h、1 d、2 d、3 d、5 d、7 d 8个亚组,每组6只。采用大鼠尾动脉自体不凝血50 μL注入尾状核区建立脑出血模型。依达拉奉治疗组于模型制作成功后2 h开始腹腔内注射3 mg/kg依达拉奉,1次/d。免疫组化法检测NF-κB p65蛋白表达变化。 结果 与脑出血组相比,依达拉奉治疗组于出血后 6 h、12 h、1 d、2 d、3 d、5 d、7 d血肿周围组织NF-κB p65蛋白表达均减少(P < 0.05或P < 0.01)。 结论 依达拉奉对脑出血后血肿周围脑组织损伤有保护作用,可能与其抑制NF-κB p65蛋白表达有关。

    Effect of Edaravone on NF-κB p65 protein expression in perihematoma of rats with intracerebral hemorrhage

    [Abstract] Objective To investigate the effect of Edaravone on the NF-κB p65 protein expression in perihematoma of rats with intracerebral hemorrhage (ICH). Methods 108 healthy adult Wistar rats were randomly divided into normal group, sham group, ICH group, Edaravon treatment group. The ICH group and Edaravon treatment group were randomly divided into 3 h, 6 h, 12 h, 1 d, 2 d, 3 d, 5 d, 7 d subgroups respectively after ICH, each group had 6 rats. ICH models were prepared by injecting 50 μL of autogenous blood into the right caudate nucleus of rats. Edaravon treatment group after ICH 2 h were given 3 mg/kg Edaravon by intraperitoneal injection, 1 time/d. The expression of NF-κB p65 protein was observed by immunohistochemistry. Results Compared with ICH group, the expression of NF-κB p65 protein in Edaravone treatment group were weakened at 6 h, 12 h, 1 d, 2 d, 3 d, 5 d and 7 d after ICH (P < 0.05 or P < 0.01). Conclusion Edaravon has a protective effect on perihematoma brain tissues after ICH, it may be related to the inhibition of NF-κB p65 protein expression. ......
