高效液相色谱—电喷雾质谱分析白杨素在大鼠体内的代谢产物
刘炜 姜金生 韦英杰[摘要] 目的 探讨白杨素在大鼠体内的代谢产物。 方法 给药组以白杨素2 mg/mL的溶液灌胃雄性SD大鼠,同时空白组大鼠以等体积的溶剂灌胃。灌胃24 h后分别取两组大鼠的尿液和粪便,同时分别于给药后0.5、1、2、4、6 h对两组大鼠进行眼眶取血。应用液质联用推测化合物的可能结构。 结果 在大鼠尿、粪便、血浆检测到原型成分白杨素M1(白杨素),在大鼠粪、胆汁中检测到M2(白杨素硫酸结合物);此外,在大鼠尿液、血、胆汁等样品中检测到代谢成分M3-M4(白杨素葡萄糖醛酸结合物)。 结论 白杨素在大鼠体内的主要代谢方式为Ⅱ相代谢,产物是葡萄糖醛酸结合物与硫酸结合物。
