哇巴因调控Akt/mTOR信号通路诱导U87—MG细胞死亡的机制研究
杨小飒 李培炎 李杰[摘要] 目的 探讨哇巴因(ouabain)对人胶质瘤U87-MG细胞死亡的影响及具体机制。 方法 将培养的U87-MG细胞随机分为正常对照(Control)组和ouabain组,分别加入常规培养基和不同浓度ouabain(0.05、0.5、2.5、25 μmol/L)干预24 h,光学显微镜观察ouabain干预后24 h细胞的形态改变,MTT实验检测细胞活力,免疫印迹法(Western blot)检测Akt、p-Akt、mTOR和p-mTOR的表达变化。 结果 与Control组比较,ouabain显著降低U87-MG细胞活力(均P < 0.01),且呈现浓度依赖性;高浓度ouabain(2.5、25 μmol/L)与Control组相比能够降低U87-MG细胞中p-Akt、mTOR和p-mTOR的表达(均P < 0.01)。 结论 高浓度ouabain可能通过抑制胞内Akt/mTOR信号通路,降低肿瘤细胞活力并最终引起细胞死亡。

[Abstract] Objective To explore the effect of ouabain on the human glioma U87-MG cells death and the underlying mechanism. Methods U87-MG cells were randomly divided into control group and ouabain group, which were treated with conventional medium and different concentrations of ouabain (0.05, 0.5, 2.5 and 25 μmol/L) for 24 h, respectively.
