外源性硫化氢对大鼠睾丸缺血再灌注损伤的保护作用
刘卫华++++++邓红琴[摘要] 目的 探讨外源性硫化氢对大鼠睾丸缺血再灌注损伤的保护作用。 方法 选取成年雄性Wistar大鼠30只随机分为3组,每组各10只:对照组(A组)、睾丸缺血再灌注损伤组(B组)、睾丸缺血再灌注损伤+硫化氢干预组(C组)。B组和C组大鼠建立睾丸扭转复位模型,C组在再灌注前腹腔注射外源性硫化氢(100 μmol/kg)。HE染色观察睾丸组织病理改变;测定丙二醛(MDA)、超氧化物歧化酶(SOD)含量;采用原位缺口末端标记检测细胞凋亡指数;免疫组化检测睾丸组织中的Bax,Bcl-2和Caspase-3的表达情况。 结果 B组可见生精上皮排列紊乱,生精细胞广泛脱落。与A组比较,B组中MDA含量和生精细胞凋亡指数明显升高,SOD活性明显下降,差异有统计学意义(P < 0.05),B组中Bax/Bcl-2比值及Caspase-3表达明显高于A组;C组明显改善睾丸缺血再灌注损伤引起的变化,C组中MDA含量、SOD活性、生精细胞凋亡指数与B组比较,差异有统计学意义(P < 0.05)。 结论 外源性硫化氢可减轻氧化应激水平和抑制生精细胞凋亡, 从而在睾丸组织在缺血再灌注损伤过程中发挥保护作用。 [关键词] 硫化氢;睾丸;缺血再灌注损伤;氧化應激;凋亡

[Abstract] Objective To investigate the protective effects of hydrogen sulfide (H2S) on the testicular ischemia-reperfusion injury in rats. Methods Thirty adult male Wistar rats were randomly divided into 3 groups, with 10 rats in each group, control group (group A), testicular ischemia-reperfusion injury group (group B), testicular ischemia reperfusion injury + H2S intervention group (group C).
