椎间隙感染后持续腰背痛的机制研究

杨欢 宋红星 崔磊[摘要] 目的 探讨金黄色葡萄球菌上清液对椎间盘髓核细胞蛋白聚糖(ACAN)、Ⅱ型胶原(COLⅡ)的影响。 方法 体外分离培养SD大鼠椎间盘髓核细胞,使用不同浓度(0%、5%、10%、15%、20%、25%、30%、35%、40%)金黄色葡萄球菌上清液处理,模拟椎间隙感染病程,采用CCK-8检测第二代髓核(P2)细胞增殖活性;采取RT-PCR检测第二代髓核细胞的基因表达量。 结果 本研究发现金黄色葡萄球菌上清液加速髓核细胞去分化,刺激ACAN、COLⅡ表达下降(P 20%时,会明显抑制髓核细胞外基质合成。 结论 经初步研究发现,金黄色葡萄球菌上清液可以抑制髓核细胞生长,且具有浓度依赖性;刺激ACAN、COLⅡ表达减少可能是造成椎间隙感染后持续腰背痛的原因。

[Abstract] Objective To Discus on the Staphylococcus aureus supernatant of intervertebral disc nucleus pulposus cells proteoglycans (ACAN), type Ⅱ collagen (COL Ⅱ). Methods Cultured SD rat intervertebral disc nucleus pulposus cells were isolated in vitro and treated with Staphylococcus aureus supernatant at different concentrations (0%, 5%, 10%, 15%, 20%, 25%, 30%, 35%, 40%) to simulate the course of intervertebral space infection. CCK-8 was used to detect the proliferation activity of the second generation nucleus pulposus (P2) cells.
