【摘要】 目的 研究猫眼草提取物对不同胃癌细胞株AGS、SGC-7901、BCG-823增殖的影响并探讨其可能的机制。方法 不同胃癌细胞株AGS、SGC-7901、BCG-823分别给予猫眼草提取物10.0、5.0、1.0、0.1 mg/L培养48 h,采用MTT法和台盼蓝染色法检测猫眼草提取物对不同胃癌细胞株增殖的影响。结果 MTT法测定结果显示,猫眼草提取物各浓度组对AGS、SGC-7901、BCG-823细胞增殖均有明显抑制作用且具有良好的量效关系(P<0.05);台盼蓝法计数结果显示,给予猫眼草提取物后,活细胞数随培养时间的延长而减少,对胃癌细胞株AGS、SGC-7901、BGC-823增殖的抑制具有良好的时效关系(P<0.05)。 结论 猫眼草提取物对胃癌细胞株AGS、SGC-7901、BCG-823增殖均具有明显的抑制作用,其抑制作用强度随给药浓度的增加和作用时间的延长而递增。

【Abstract】 Objective To investigate the effects and mechanism of Maoyancao extract on gastric cancer cell line AGS、SGC-7901、BCG-823.Methods Gastric cancer cell line AGS、SGC-7901、BCG-823 were treated with Maoyancao extract of different concentration or different time and effect of Maoyancao extract on proliferation of AGS、SGC-7901、BCG-823 was evaluated by MTT and trypan-blue assay.Results Determination of MTT results showed that different concentration ofMaoyancao extract has significant inhibitory effect on gastric cancer cell line(P<0.05)and the inhibitory effect increases with the increase of concentration;determination of MTT results showed that with the increase of time living cells decrease,Maoyancao extract has significant inhibitory effect on gastric cancer cell line AGS,SGC-7901,BCG-823.Determination of MTT results showed that.Conclusion The Maoyancao extract has significant inhibitory effect on gastric cancer cell line AGS,SGC-7901,BCG-823and the inhibitory effect increases with the increase of concentration and time.

猫眼草又名泽漆,是大戟科植物猫眼草的全草,性微寒,味苦,有毒。在民间曾有用单味猫儿眼生药治疗胃癌、食道癌的记载[1]。多项研究也已发现猫眼草中含有黄酮类等抗癌成分[2,3],但相关文献资料对猫眼草的抗胃癌作用机制研究甚少,本实验选用猫眼草制得提取物,对人不同胃癌细胞株AGS、SGC-7901、BCG-823增殖的影响进行实验研究,为猫眼草的开发研究和临床应用提供科学依据。
