血管紧张素Ⅱ受体拮抗剂洛沙坦对脓毒症大鼠多脏器的保护作用(1)

     [摘要]目的 探讨血管紧张素Ⅱ受体(AT1型)特异性拮抗剂洛沙坦(losartan)对脓毒症大鼠心、肝、肾、肺、小肠等多脏器的保护作用及对预后的影响。方法 SD清洁级大鼠120只,雌雄不拘,随机分为4组,即假手术组(30只)、脓毒症组(30只)和脓毒症大鼠30mg/kg剂量的洛沙组在制模前预给药、制模后给药造成处理组(各30只)。采用改良盲肠结扎穿孔方法(CLP)建立大鼠脓毒症模型。观测各组动物心、肝、肾、肺、小肠等脏器功能指标。各组剩15只大鼠计算5d累计死亡率。结果 与假手术组比较,脓毒症组大鼠心、肝、肾、肺、小肠等功能指标有显著变化 (P<0.05),病理损害较严重,死亡率显著升高(P<0.05);与脓毒症组比较,给予洛沙组处理的两组大鼠心、肝、肾、肺、小肠等功能指标与脓毒症组比较显著好转(P<0.05),病理损害较轻,死亡率显著下降(P<0.05)。两洛沙坦处理组无明显差异(P>0.05)。结论 洛沙坦对脓毒症大鼠心、肝、肾、肺、小肠等多脏器具有保护作用,并能改善预后。

    [Abstract] Objective To study the role of Angiotensin Ⅱreceptor (AT1-type) antagonist losartan in rat with sepsis on acute protective effect of multiple organs including heart,liver,kidney,lung and intestine. Methods One hundred and Twenty SD rats were randomly divided in to sham operated group(SHAM), septic group(CLP), losartan pre-CLP group (pre-CLP) and losartan post-CLP group(pre-CLP).Each group has 30 rats.Sepsis model was produced by cecalligation and puncture(CLP). To observe the organ function parameters in all groups of heart,liver,kidney,lung and intestine.Remaining 15 rats in each group calculated the 5d cumulative mortality rate.Results Compared with SHAM group,the organ function parameters of CLP group were significantly abnormal(P<0.05),pathological damage were more severe, the Mortality rate were significantly increased(P<0.05). Compared with CLP group, the organ function parameters of the two losartan groups were Significantly improved (P<0.05),the Mortality rate were significantly decreased(P<0.05),and had Less pathological damage.
