[摘要] 目的:研究林可霉素对于健康人外周血单核巨噬细胞表面分子表达的影响,探讨林可霉素对单核巨噬细胞免疫学功能的影响。方法:从健康人外周血中提取单核巨噬细胞体外培养,培养7 d后将其分为霉素处理组和对照组,林可霉素处理组加入林可霉素,对照组加入0.9%氯化钠溶液,刺激48 h后,流式细胞仪检测细胞表面CD80、MHCⅡ及Fas表达,ELISA法检测细胞培养上清液中细胞因子人干扰素-γ(IFN-γ)、肿瘤坏死因子α的浓度。结果:在林可霉素处理后,单核巨噬细胞抗原呈递相关分子CD80、 MHCⅡ表达降低,细胞因子IFN-γ、TNF-α表达明显减少,其表面死亡受体Fas表达升高。结论:林可霉素可明显抑制单核巨噬细胞抗原呈递及细胞因子分泌等免疫功能,并诱导单核巨噬细胞死亡。

The effect of lincomycin on the immune function of human peripheral blood-derived mononuclear macrophage cells in vitro

[Abstract] Objective: To investigate the effect of lincomycin on the surface molecules expression level of human peripheral blood-derived mononuclear macrophage cells in vitro. Methods: Macrophage cells were extracted in vitro from peripheral blood of health volunteers and incubated with lincomycin for 48 h, the variation of CD80, MHCⅡ and Fas were detected with flow cytometric analysis, and the secretion of IFN-γ and TNF-α by macrophage cells were detected with ELISA. Results: The lincomycin decreased the expression of antigen-presenting factor CD80, MHCⅡ of macrophage cells and their production of IFN-γ, TNF-α, and increased the Fas expression on the cells. Conclusion: Lincomycin inhibits immune function of the macrophage cells, and induces their death.

林可霉素是一种临床常用药,对G+菌及部分厌氧菌有明显杀菌作用,然而,目前发现其对机体整体免疫功能有明显的抑制作用[1]。本研究针对免疫系统中起关键作用的单核巨噬细胞进行研究,从健康人外周血培养骨髓DC,经林可霉素刺激48 h,探讨其对小鼠DC的表面共刺激分子以及免疫功能的影响。
1 材料与方法
1.1 材料及主要试剂
健康人外周血来自于20~25岁健康志愿者;人重组M-CSF购自Peprotech,PE标记的鼠抗人MHCⅡ(HLA-DR)抗体(同型对照为PE标记小鼠IgG2a),PE-鼠抗人CD80(同型对照为PE标记小鼠IgG2b),人干扰素-γ(IFN-γ),肿瘤坏死因子α(TNF-α)细胞因子,ELISA检测试剂盒均购自美国ebioscience公司;PE标记鼠抗人Fas(同型对照为PE标记小鼠IgG1)购自美国Biolegend公司;完全培养基RPMI1640购自美国hyclone公司;胎牛血清购自Gibco公司。
