岩大戟内酯B诱导乳腺癌MCF-7细胞凋亡的实验研究(1)

【摘要】 目的：探讨岩大戟内酯B对MCF-7乳腺癌细胞生长抑制作用及其机理。方法：通过MTT法观察各组细胞增殖能力；采用流式检测技术分析细胞周期；利用Annexin V/PI双染检测细胞凋亡情况；电镜观察细胞形态。结果：随着岩大戟内酯B浓度增加对MCF-7的抑瘤率增高；G2/M期细胞减少，S期细胞增多，出现S期阻滞；晚期凋亡和死亡的细胞增加；电镜显示加药后细胞出现核固缩，细胞器空泡变性，凋亡小体形成，甚至出现坏死。结论：岩大戟内酯B对乳腺癌MCF-7细胞生长明显抑制作用，并诱导肿瘤细胞凋亡，为临床开发新药提供理论依据。

【Abstract】 Objective: To investigate the inhibition effects of Jolkinolide B on breast cancer cell growth inhibition and its mechanism. Methods: Author used MTT method to observe the proliferation of MCF-7 cell lines when treat with Jolkinolide B; Cell cycle was assayed by flow cytometer; Annexin V/PI double dye was detected to cell apoptosis; Cell morphology was observed by electronic microscopy. Results: The inhibit ratios of MCF-7 cell lines were increased following the drug concentrations of Jolkinolide B; The G2 / M period cells reduce， S phase increased the number of cells and increase apoptosis rates， it appears the block of S phase; There were a lot of apoptotic cell and dead cell; Many nuclei had become pyknotic by electronic microscopy， organelles empty bubble degeneration， appear apoptotic body， even necrosis after dosing the different concentration of drug. Conclusion: Jolkinolide B can significantly inhibit the growth of MCF-7 breast cancer cell and induced apoptosis. It is provides the theory for a new drug with the development of clinical therapy.

狼毒大戟(euphorbia fischeriana steud)含二萜醇类化合物，其化学成分主要包括萜类、鞣质类、甾醇类、挥发油成分等。包括岩大戟内酯(jolkinolide)A、B，狼毒大戟甲、乙素(fischeriana A，B)等，并含(euphol)、皂甙、强心甙、甾醇、酚类及鞣质。散播结，杀虫。外用于淋巴结结核、皮癣；灭蛆。 治疗肺癌、肝癌、胃癌等恶性肿瘤临床疗效显著，未发现对心、肝、肾等造血系统的不良反应[1-2]。不同化学方法提取中药单体成分的研究较为深入[3-5]。本文仅对岩大戟内酯B对MCF-7乳腺癌细胞抑瘤作用进行探索，为以后临床治疗提供实验依据。
1 材料与方法
1.1 细胞系 MCF-7乳腺癌细胞由北京大学肿瘤医院肿瘤分子生物学研究室惠赠。
1.2 主要仪器和药物 岩大戟内酯B由齐齐哈尔大学帮助提取，主要试剂DMEM培养基购自美国Gibco公司，胎牛血清购自美国Hyclone ......
