急性胰腺炎时腺泡细胞凋亡与Bcl-2表达(1)

     【摘要】 目的: 探讨急性胰腺炎(AP)时腺泡细胞凋亡与Bcl-2表达的关系。方法: SD大鼠30只,随机分为假手术组(A组),急性水肿性胰腺炎组(B组),急性出血坏死性胰腺炎组(C组)。采用逆行胰胆管穿刺注射法建立急性水肿性与出血坏死性胰腺炎模型。检测造模后12 h各组的血清淀粉酶活性、腹水情况、胰腺组织大体及镜下病理改变与评分、腺泡细胞凋亡指数(AI)、Bcl-2蛋白表达情况。结果: ⑴ C组血清淀粉酶活性、腹水量、大体及镜下病理评分显著高于B组(P<0.05),Bcl-2阳性率、腺泡细胞AI显著低于B组(P<0.05)。⑵ B、C组中腺泡细胞AI与大体及镜下病理评分均呈显著负相关关系(P<0.05)。结论: AP时腺泡细胞的凋亡是机体的一种自我保护机制,Bcl-2参与了腺泡细胞凋亡的调控。

    【Abstract】 Objective:To explore the relationship between apoptosis of acinar cells and Bcl-2 expression in rats with acute pancreatitis.Methods:Thirty SD rats were randomly divided into three groups: group A(shame operation group),group B(acute edematous pancreatitis group)and group C(acute hemorrhagic necrotic group).Two kinds of pancreatitis model were induced by retrogradely injection of sodium deoxycholate.After 12 hours,the serum amylase activity,ascites,apoptosis of acinar cells and expression of Bcl-2 were measured,the general and histological changes of pancreas were evaluated by the corresponding score system.Results:⑴The serum amylase activity,ascites volume,general and histological pathology score in group C were significantly higher than group B(P<0.05),positive rate of Bcl-2 and AI of acinar cells were lower than group B(P<0.05).⑵ In group B and C,AI were negatively correlated with general score and histological score(P<0.05).Conclusion:The apoptosis of acinar cells is a self-protection mechanism of individuals with AP,Bcl-2 is involved with the regulation of the acinar cells apoptosis.

    急性胰腺炎(acute pancreatitis,AP)是常见的外科急腹症,其发病机制尚未完全阐明。最近发现,胰腺腺泡细胞凋亡与AP的发生、发展及转归密切相关,使之成为当前的研究热点。Bcl-2是广受关注的凋亡相关基因,可促进细胞的凋亡,但目前国内外关于Bcl-2与AP时腺泡细胞凋亡的研究鲜有报道。本研究旨在探讨AP时腺泡细胞凋亡与Bcl-2表达的关系。
