乌司他丁预处理对创伤性颅脑损伤大鼠神经保护作用的初步研究(1)

     【摘要】 目的:探讨乌司他丁(UTI)预处理对SD大鼠颅脑损伤早期的神经保护作用及其机制。方法:选取雄性SD大鼠48只,随机分为假手术组(SO组)、乌司他丁预处理组(UTI组)、生理盐水对照组(Control组)和创伤性脑损伤组(TBI组)。分别给予UTI组和Control组连续3 d UTI 300 000 U/(kg·d)及等体积生理盐水腹腔注射。除SO组外,其余各组采用自由落体打击法建立创伤性脑损伤模型。打击后,采用改良的神经功能缺损评分评估四组大鼠的神经功能,干湿重法检测脑组织含水量,连续切片行HE染色后计算脑损伤的体积,经尾静脉采血用ELISA法检测血清S100B蛋白及NSE浓度变化。结果:UTI组颅脑损伤早期神经功能缺损评分、脑组织含水量及脑组织损伤体积均小于Control组(P<0.05),同时UTI组的血清S100B蛋白及NSE浓度均低于Control组(P<0.05),且UTI组、Control組及TBI组三组的血清S100B蛋白及NSE浓度均呈双峰变化。结论:乌司他丁预处理可降低实验大鼠颅脑损伤早期血清S100B蛋白及NSE浓度,减轻脑水肿及脑组织损伤体积,改善神经功能缺损,具有神经保护作用。

    【Abstract】 Objective:To investigate the protective mechanism of Ulinastatin on traumatic brain injury rat model.Method:Forty-eight SD rats were randomly divided into four groups:sham operation group(SO group),NaCl control group(Control group),UTI pretreatment group(UTI group),and traumatic brain injury group(TBI group).UTI group and Control group were given UTI 300 000 U/(kg·d) and the same volume of saline by intraperitoneal injection respectively.A closed TBI model was established by weight-drop device in the other groups except for SO group.The modified criteria of neurological defecting score was adopted to evaluate the nerve function of four groups.The wet-dry weighing method was used to detect the brain water content.The volume of brain injury was calculated after HE staining with serial section.The serum S100B protein and NSE from tail vein were detected by ELISA.Result:Compared with Control group,the score of neurological impairment,the water content,the volume of brain tissue defecting of UTI group were less(P<0.05).The serum S100B protein and NSE concentration of UTI group were lower than those of Control group(P<0.05).In addition,the serum S100B protein and NSE concentration in UTI group,Control group and TBI group showed a double peak change.Conclusion:Ulinastatin pretreatment will decrease the serum S100B protein and NSE concentration of the rats with traumatic brain injury in early stage,reduce cerebral edema,decrease the volume of brain tissue injury,improve nerve function defect and has a nerve protective effect.
