金福菇多糖对衰老大鼠p16基因表达影响(1)
【摘要】 目的:探讨金福菇多糖对衰老大鼠p16基因表达的影响。方法:实验材料选用清洁级SD大鼠25只,将实验动物随机分成5组,每组5只。分别为正常对照组、衰老模型组、高剂量治疗组、中剂量治疗组、低剂量治疗组,采用皮下注射D-半乳糖方法建立衰老大鼠模型,除正常对照组外,分别给予金福菇多糖溶液灌胃治疗,采用逆转录PCR(reverse transcription-PCR,RT-PCR)检测各组大鼠心脏组织p16 mRNA的表达情况。结果:衰老模型组小鼠p16基因表达明显高于正常对照组,差异有统计学意义(P<0.05),给予金福菇多糖治疗后p16基因相对表达均明显低于衰老模型组小鼠,其中中剂量组最低,三组比较差无统计学意义(P>0.05)。结论:金福菇多糖可有效降低p16 mRNA的表达,从而起到抗衰老的作用,值得进一步研究。

【Abstract】 Objective:To explore the effect of Jin Fu Mushroom Polysaccharide on p16’s gene expression of aging rats.Method:A total of 25 clean grade SD rats were chosen,the animals were randomly divided into 5 groups,5 rats in each group(the normal control group,aging model group,high-dose treatment group,middle-dose group,small-dose treatment group),using the method of subcutaneous injection of D-galactose aging rat model was established,in addition to the normal control group,another groups were given different doses of Jin Fu Mushroom Polysaccharide treatment,the p16 gene expression of rats was detected by reverse transcription PCR(reverse transcription-PCR,RT-PCR).Result:The p16 gene expression of aging model group was obviously higher than that of the normal control group,the difference was statistically significant(P<0.05),after giving Jin Fu mushroom polysaccharide,the p16’s gene expression was lower than those of in aging model group,in which the lowest dose group,but there was no significant difference among three groups(P>0.05).Conclusion:Jin Fu Mushroom Polysaccharide can effectively reduce the p16 mRNA expression,is likely to be the mechanism of heart in the rat model of aging,is worth of further study.

隨着生物医学的发展,人们对食用菌多糖及其衍生物的活性有了更深入的了解,发现食用菌多糖具有一定的辅助医疗作用,如抗菌、抗病毒、抵抗缺氧环境、抗衰老、抗肿瘤、降血脂等,是保健性食品及抑癌药物的重要原料来源[1-3],被专业人士所认可。近年来,国内外关于食用菌研究很多,但多局限于食用菌培养条件的优化及其生物学特性[4-5],而关于金福菇多糖抗衰老的研究较少。衰老与基因有密切关系,被某些基因所控制,随着年龄的增加,多数原癌基因表达或活性降低,使得机体组织和器官发生渐进性退行性变,直到死亡,这是所有生物的特征[6]。细胞衰老是器官功能乃至整个机体功能衰老的主要表现[7]。p16基因是一种抑癌基因,在细胞衰老过程中起着重要的作用[8]。研究表明p16基因作用诱导细胞老化,抑制细胞无限分裂,从而维持细胞周期稳态[9-10],金福菇是一种分布于热带和亚热带的食用菌,具有丰富营养价值,且具有多种生物活性,近年来,多个课题组成员对金福菇多糖及生物活性进行研究,金福茹多糖具有很好的抗氧化活性[11-12]。金福菇多糖具有抗氧化活性、抗肿瘤、抗疲劳、调节免疫等作用[13]。本实验探讨金福菇多糖对衰老大鼠p16基因表达的影响,从而能够抑制大鼠衰老的作用,旨在对金福菇多糖抗衰老的生物学活性进行研究,为抗衰老的药物研究提供可靠依据。现报道如下。
1 材料与方法
