【摘要】 p53基因是重要的抑癌基因之一，具有抑制肿瘤的作用，超过50%的癌症伴有p53基因的突变。p53基因的突变不仅导致其肿瘤抑制功能的失去，同时将促进肿瘤的发生发展，此现象被称为获得性新功能(GOF)。本文将从p53与细胞周期、细胞代谢途径及干扰转录调控等方面综合阐述p53与癌症之间的关系，为癌症的创新治疗及靶向药物的开发提供理论依据。
